Determination of lignans and related substances

Abstract:

V révě vinné, víně, ale i dřevě byl prokázán obsah lignanů, látek patří-cích do skupiny sekundárních metabolitů. Jsou to látky, které zastávají důležitou roli v obraně rostliny, procesech fotosyntézy, dýchání a mnoha dalších. V rámci pozitivního efektu na lidské zdraví hovoříme o správné činnosti kardiovaskulárního systému, kognitivních funkcích, zdraví kostí, prevenci proti rakovině a potlačení menopauzálních symptomů. Cílem této práce není pouhé stanovení koncentrace lignanů ve víně. Nýbrž i srovnání koncentrace těchto látek ve víně zrajícím v nerezovém tanku, dubovém sudu a hroznovém moštu, ze kterého bylo víno vyrobeno. Dále pak určení původu lignanů ve víně. V rámci práce byla provedena optimalizace metod stanovení lignanů pomocí LC/DAD a LC/MS. Vybranými zástupci lignanů jsou isolariciresinol, pinoresinol a matairesinol. V hroznovém moštu byl detekován pouze matairesinol. Ve víně potom isolariciresinol, pinoresinol a matairesinol ve zvýšené koncentraci. Rozdíl koncentrace lignanů ve víně zrajícím v nerezovém sudu a dřevěném tanku nebyl potvrzen. Byla zaznamenána pouze nižší koncentrace isolariciresinolu ve starším a vícekrát použitém dubovém sudu. Na základě provedeného měření je původ lignanů v námi zkoumaném víně přisuzován vyluhováním z peciček.

Abstract:

The content of lignans, substances belonging to the group of secondary metabolites, was proved in vines, wine, but also wood. They are substances that play an important role in plant defence, the processes of photosynthesis, respiration and many others. As part of the positive effect on human health, we talk about the proper functioning of the cardiovascular system, cognitive functions, bone health, cancer prevention and suppression of menopausal symptoms. The aim of this work is not just to determine the concentration of lignans in wine. There is also a comparison of the concentration of these substances in the wine aged in stainless steel, in oak barrels and the grape must which the wine was made from. Furthermore, determining the origin of lignans in wine. Within the work optimization of lignan determination by LC/DAD and LC/MS was created. Isolariciresinol, pinoresinol and matairesinol are selected representatives of lignans for this measurement. Only matairesinol was detected in grape must. Isolariciresinol, pinoresinol and matairesinol in higher concentration were ascertained in wine. The difference in the concentration of lignans in the wine aged in a stainless steel barrel and a wooden tank was not confirmed. Only lower concentrations of isolariciresinol was found in older and reusable oak barrels. Based on the performed measurement, the origin of the lignans in the wine we were researching is attributed to leaching from the kilns.
