Options of inhibition of the mitotic spindle by modern medicaments in the blood cancer issue

Abstract:

Cílem této bakalářské práce je přiblížit jakémukoliv čtenáři rakovinovou problematiku, s níž se čím dál častěji setkáváme. To zahrnuje jak pochopení buněčného cyklu jako celku, tak i jeho jednotlivých fází (interfáze a M-fáze). Důležitou částí jsou také regulace buněčného cyklu, které zahrnují kombinaci stimulačních a inhibičních faktorů. Právě tato kombinace faktorů ovlivňuje části buněčného cyklu a řídí důležité procesy replikace DNA, mitózu i cytokinezi. Za normálních okolností regulační faktory zabraňují vzniku nádorového bujení. Pokud ovšem dojde ke změnám, způsobených určitými vlivy (vnějších a genetických), může docházet k přeměně normální buňky v buňku nádorovou. Vnější faktory patří mezi nejvýznamnější, jelikož mohou ovlivňovat vznik rakoviny až ze 75-95% všech nádorů. V případě vzniku rakoviny, mohou ovlivnit, zda se bude jednat o zhoubné či nezhoubné rakovinové bujení. Rizikové faktory jsou zejména kouření a strava, nadváha a obezita, alkohol, nedostatek pohybu a infekce. Mezi genetické vlivy jsou řazeny zejména mutace buněk, které byly vyloučené z regulačních procesů buněčného cyklu. A díky tomu se dnes nedíváme na rakovinu jako na jedno onemocnění, ale jako skupinu chorob. Což umožňuje komplexnější a cílenější léčbu. V poslední řadě jsem se snažila propojit vysvětlení procesu mitózy ve spojení s inhibitorem ARRY-520, jež zapříčiňuje právě zastavení tohoto procesu u rakovinných buněk a tím zabraňuje dalšímu dělení takto nemocných buněk.

Abstract:

The main object of this bachelor?s work is to give everybody insight into cancerous problems, which are becomming more common. It is inclusive of understanding the cell cycle as a whole, as well as every single phase (interphase and the M-phase). An important part are also the regulations of the cell cycle, which include the combination of stimulation and inhibitory factors. Just this combination of factors affects the cell cycle parts and controls the important processes of DNA replications, mitosis and cytokinesis. Under normal circumstances, the regulation factors prevent the formation of tumorous growth. However, if there are changes, caused by certain influences (external and genetic), it may comes to transformation of the normal cell into the tumor cell. The external factors belong to the most important, because they can affect the start of cancer up to 75-95 % of all tumor. In the case of formation of cancer, the factors may affect, whether it will be about a melignant tumor or a benign tumor. High-risk factors include especially smoking and diet, overweight and obesity, alcohol, activity deficiency and infection. Within the genetic influences belong especially the mutation of cells, which were excluded from regulation processes of the cell cycle. And because of that we don?t look at the cancer as at only single disease, but we look at it as a group of diseases. Which enables more comprehensive and targeted treatment. Finally, I have tried to connect the processes explenation of mitosis in uniting with inhibitor ARRY-520, which causes just stopping of this process by the cancer cells, thus prevents to further separation of the cells diseased like this.
