The role of transgelin in renal cell carcinoma development

Abstract:

Transgeliny jsou proteiny vázající aktin a tím jsou nepřímo zapojeny do mnoha buněčných procesů, jako je buněčná migrace, proliferace, diferenciace a apoptóza. V rešerši práce je popsána struktura, exprese a biologická funkce této rodiny, do níž patří transgelin a jeho homology trangelin -2 a -3 s různou tkáňovou distribucí. Mnoho studií popisuje deregulaci exprese transgelinu v různých typech nádorů, což může ovlivňovat jejich vývoj. Změněná hladina transgelinu byla pozorována také u karcinomů ledvin špatně odpovídajících na léčbu sunitinibem v naší pilotní proteomické studii z roku 2018. V práci je proto stručně popsána biologie karcinomu ledviny a aktuální terapeutické postupy. Cílem práce bylo popsat vliv změn v hladinách transgelinu na buněčnou proliferaci a migraci. Byly připraveny buněčné linie odvozené z renálního karcinomu s umlčeným genem pro transgelin pomocí metody CRISPR/Cas9 bez selekce pozitivně transfekovaných buněk. Buněčná linie 786-0 byla úspěšně transfekována a k analýze hladiny transgelinu pomocí SDS-PAGE a imunoblotingu narostlo celkem 42 klonů. Z těchto klonů bylo 23 klonů s nezměněnou hladinou, 8 se sníženou hladinou a 9 s nízkou hladinou transgelinu. U klonů s nízkou hladinou byla provedena sekvenace DNA u 5 klonů a modifikace genu pro transgelin nebyla pozorována; 3 klony vymřely během kultivace před analýzou DNA. Ani u jednoho klonu nedošlo k úplnému umlčení exprese transgelinu. U všech klonů byla sledována rychlost proliferace. Z výsledků vyplývá, že klony s nízkou hladinou transgelinu rostou pomaleji než klony s nezměněnou nebo sníženou hladinou transgelinu. Při použití systému CRISPR/Cas9 se selekcí pozitivně transfekovaných buněk nebyly u linie 786-0 pozorovány přežívající buňky, zatímco u linie CAKI-1 bylo vyselektováno na 40 klonů při stejném postupu. Celkově lze říci, že transgelin je pravděpodobně pro linii 786-0 esenciální. Dalším přístupem pro sledování role transgelinu při migraci bylo jeho transientní umlčení metodou RNA interfence pomocí siRNA. Pilotní pokus u linie CAKI-1 nevedl ke snížení hladiny transgelinu a je nutná další optimalizace tohoto přístupu.

Abstract:

Transgelins are actin-binding proteins and are thus indirectly involved in many cellular processes, such as cell migration, proliferation, differentiation and apoptosis. The research describes the structure, expression and biological function of this family, which includes transgelin and its homologues trangelin -2 and -3 with different tissue distribution. Many studies describe the deregulation of transgelin expression in various types of tumors, which may influence their development. Altered transgelin levels were also observed in renal cell carcinomas poorly responding to sunitinib treatment in our pilot proteomic study in 2018. The biology of renal cell carcinoma and current therapeutic approaches are therefore briefly described in this work. The aim of this work was to describe the effect of changes in transgelin levels on cell proliferation and migration. Renal carcinoma-derived cell lines with silenced transgelin gene were prepared using the CRISPR/Cas9 method. The 786-0 cell line was successfully transfected to give a total of 42 clones. These clones were analyzed by SDS-PAGE followed by immunodetection, and the analysis showed that 23 clones were unchanged, 8 had reduced transgelin, and 9 had low transgelin level. Sequencing of DNA for TAGLN exon 2 were done in 5 clones with low transgelin level and DNA modification was not found. 3 clones with low transgelin level died before DNA analysis. There was not any clone with completely silenced transgelin expression. All clones were observed for proliferation rate. Based on the results, the clones with low trangelin level grew slower than clones with unchanged or reduced transgelin level. There was not any cell survived in CRISPR/Cas9 system with selection of positively transfected cells in 786-0 line, while about 40 clones were selected in CAKI-1 line. All together, trangelin is probably essential for 786-0 cells. Another approach how to observe the role of transgelin in migration is RNA interference using siRNA. Pilot experiment in CAKI-1 cells did not lead to transgelin silencing and further optimization of the process in necessary.
