Studium interakcí homologních transgenních lokusů v hybridních rostlinách Nicotiana tabacum

Abstract:

Tato práce se zabývala studiem meiotické a mitotické stability epigenetického stavu transgenních lokusů v hybridních rostlinách N. tabacum a v tkáňových kulturách z těchto rostlin odvozených. Hybridní rostliny obsahovaly posttranskripčně nebo transkripčně umlčený lokus 1 uspořádaný jako obrácené opakování, v kombinaci s aktivně exprimovaným homologním transgením lokusem. Na základě expresních a metylačních analýz bylo zjištěno, že během dvou resp. tří generací je epigenetický stav transgenních lokusů v rostlinách Lo1/Lo2 (kombinují posttranskripčně umlčený lokus 1 a aktivní lokus 2) a Lo1E/Lo2 (transkripčně umlčený lokus 1 a aktivní lokus 2) stabilně udržován. Posttranskripčně umlčený lokus 1 je schopen i v hybridech Lo1/Lo2 druhé generace komunikovat s homologním transgenním lokusem a in trans navodit jeho metylaci a umlčení. Transkripčně umlčený lokus 1E této interakce schopen není, lokus 2 zůstává v hybridech Lo1E/Lo2 nemetylován a aktivně exprimován i ve druhé a třetí generaci. V hybridních rostlinách tedy v průběhu meiózy nedochází k epigenetickým změnám, které by ovlivňovaly povahu interakcí mezi homologními transgenními lokusy. Pro definitivní zodpovězení otázky, zda-li je příslušný epigenetický stav dlouhodobě stabilně děděn, by musely být připraveny další generace hybridních rostlin. Expresní a metylační analýzy byly provedeny i u tkáňových kultur založených z hybridních rostlin Lo1/Lo2 a Lo1E/Lo2. Byla pozorována tendence buněk kalusových kultur efektivně zabránit expresi transgenních lokusů. Oba typy tkáňových kultur se ustálily ve stavu, kdy posttranskripčně umlčená varianta lokusu 1 in trans indukuje umlčení reportérového genu homologního transgenního lokusu 2. Již po třech měsících kultivace kalusové kultury Lo1E/Lo2 byla detekována hypometylace promotoru transkripčně umlčené varianty lokusu 1, která byla postačující k obnově jeho transkripční aktivity a tedy schopnosti indukovat umlčení homologního lokusu 2. Nově navozený epigenetický stav byl dále v kalusové kultuře Lo1E/Lo2 stabilně udržován. Linie Lo1/Lo2 byla během roční kultivace v tkáňové kultuře stabilní, neměnil se metylační profil transgenních lokusů a exprese reportérového transgenu zůstala na nízké úrovni srovnatelné s mateřskou rostlinou.

Abstract:

This work was focused on the study of meiotic and mitotic stability of epigenetic state of transgenic loci in hybrid plants Nicotiana tabacum and in derived cell cultures, respectively. The hybrid plants carried either posttranscriptionally or transcriptionally silenced transgenic locus 1 (arranged as inverted repeat), and an actively expressed homologous transgenic locus (locus 2). Stable reporter transgene expression and T-DNA methylation was revealed in both types of hybrid plants, Lo1/Lo2 (combining posttranscriptionally silenced locus 1 and active locus 2) and Lo1E/Lo2 (combining transcriptionally silenced locus 1E and active locus 2) over generations. Posttranscriptionally silenced locus 1 was shown to be able of communication with the homologous active locus in the second generation of hybrids and in trans induce its methylation and silencing. Transcriptionally silenced epiallele 1E failed to exhibit these interactions, locus 2 was found to be stably expressed and non-methylated in the second and third generations of Lo1E/Lo2 hybrids. Based on these results it can be concluded that no epigenetic changes possibly affecting interactions between homologous transgenic loci were detected during meiosis. Next generations of hybrid plants have to be analyzed for definite elucidation of inheritance of epigenetic pattern. Strikingly, in cell cultures derived from the Lo1/Lo2 and Lo1E/Lo2 hybrid plants trend to silence expression of the reporter gene settled in transgenic loci was observed. Both types of the callus cultures came to the state of in trans silencing of the locus 2 reporter transgene expression by the posttranscriptionally silenced locus 1. During three months of cultivation, hypomethylation of the promoter sequence of the locus 1E in Lo1E/Lo2 hybrid leading to the restoration of its in trans silencing properties was installed. New epigenetic state has been stably propagated for at least one year. Lo1/Lo2 transgenic line was stable (concerning methylation status of both transgenic loci and reporter trangene expression) during one-year cultivation in cell culture.
