Processing and archiving of microscope slides of monogenean parasites of the genus Gyrodactylus, creating of checklist of monogeneans of freshwater fish based on the research activities of department of Parasitology in Brno

Abstract:

Tato práce je zaměřena na shrnutí problematiky metodik fixace monogeneí, a dále pak fotodokumentace, měření determinačně významných struktur a tvorbu databáze, která se stane podkladem pro checklist monogeneí získaných z výzkumných aktivit oddělení parazitologie. Základem archivace je převádění dočasných preparátů na trvalé a jejich uchování, jenž poslouží pro zlepšení orientace v získaném materiálu pro potřeby budoucího srovnávání a determinace. Cílovou skupinou jsou zástupci třídy Monogenea (Platyhelminthes), a to zejména živorodé druhy rodu Gyrodactylus (Monogenea: Gyrodactylidae). V první časti se hovoří o biologii, životním cyklu, distribuci studované skupiny parazitů a také o patogenním vlivu rodu Gyrodactylus. Dále se práce věnuje vybraným metodám zpracování získaného materiálu na pozorovatelné preparáty, ať už pro studie biochemické, morfometrické, molekulární, histologické, či imunologické. Zmíněny jsou různé možnosti fixování, a to podle potřeb studia materiálu. Větší pozornost je věnována metodice, jenž je používána pro účely determinace, a to převádění preparátů z gylcerin amonium pikrátu (GAPu) do kanadského balzámu.

Abstract:

This work is focused on the issue of summary methodologies fixation Monogenea, and then photographs, measurements major structures of determination and the creation of a database that becomes the basis for Monogenea checklist obtained from research activities department of parasitology. The basis for filing is transfering temporary, and preparations for permanent preservation, which will serve to improve orientation in obtaining material for the purposes of future comparison and determination. The target groups are representatives of Monogenea (Platyhelminthes), especially viviparous species of the genus Gyrodactylus (Monogenea: Gyrodactylidae). The first part talks about the biology, life cycle, distribution of study group of parasites and the pathogenic impact of the genus Gyrodactylus. Furthermore, the work deals with the selected processing methods obtained for observable material preparations, whether for studies of biochemical, morphometric, molecular, histological, or immunological. Mentioned are the various options fixing, according to the needs of the study material. Greater attention is paid to methodology, which is used for determination, and to transfer slides from ammonium picrate-gylcerine (GAP) in Canadian balsam.
