Synthesis, Characterization and Evaluation of the Cytostatic Activity of Vanadyl-based Complexes Containing N,N-chelating Ligands

Abstract:

Diplomová práce je zaměřena na přípravu vanadylových komplexů s N,N-chelátujícími ligandy, jejich charakterizaci analytickými a spektroskopickými metodami a stanovení inhibiční aktivity IC50 pomocí WST-1 testu. Teoretická část zahrnuje obecné informace o vanadu, vanadylových sloučenin, medicinální využití a toxicitu cytostatických látek až po metody hodnocení cytotoxicity in vitro. Praktická část popisuje přípravu 9 komplexů, která spočívá v reakci vanadyltoluen sulfonátu s příslušnými ligandy (bipy, phen, ph2phen, bPM, Me2bipy a (OMe)2bipy) za vzniku těchto komplexů VO1-VO9. Průběh reakce komplexů byl sledován pomocí EPR spektroskopie. V případě komplexů VO5, VO6 a VO9 byla navržená struktura potvrzena RTG analýzou. Inhibiční aktivita 9 vanadylových komplexů byla testována pomocí WST-1 testu na buněčných linií MOLT-4 a Jurkat.

Abstract:

This diploma thesis is focused on the preparation of vanadyl complexes with N, N-chelating ligands, their characterization by analytical and spectroscopic methods and determination of IC50 inhibitory activity using the WST-1 assay. The teoretical part includes basic information about vanadium, vanadyl compounds, medical use and toxicity of cytostatic substances to methods of in vitro cytotoxicity assessment. The practical part describes the preparation of 9 complexes, which consists in the reaction of vanadyltoluene sulfonate with the appropriate ligands (bipy, phen, ph2phen, bPM, Me2bipy a (OMe)2bipy) to form these VO1-VO9 complexes. The process was studied by EPR spectroscopy. The proposed structures of complexes VO5, VO6 and VO9 were confirmed by X-ray analysis. The inhibitory activity of 9 vanadyl complexes was tested using the WST-1 assay on MOLT-4 and Jurkat cell lines.
