The use of the comet assay for the evaluation of the genotoxicity

Abstract:

V této diplomové práci jsou srovnávány genotoxické vlastnosti peroxidu vodíku a manganistanu draselného pomocí kometového testu. Byla porovnávána aplikace vzorku na buňky v médiu a aplikace na buňky zalité v agarózovém gelu. Vzorky se aplikovaly na buněčné linie HaCaT a A549. Procentuální zastoupení DNA v ocasech komet bylo vyhodnocováno softwary CASP a LUCIA Comet Assay. Tyto faktory byly testovány, zda mají vliv na míru poškození DNA v ocasech komet. Při aplikaci peroxidu vodíku na buňky byl zjištěn vliv aplikované koncentrace peroxidu, způsobu aplikace, buněčné linie i výběru softwaru. Při aplikaci manganistanu draselného byl zjištěn vliv koncentrace manganistanu a výběr softwaru pouze při aplikaci vzorku na buňky v médiu. Byly zjištěny rozdíly v poškození DNA mezi použitými buněčnými liniemi. Vliv způsobu aplikace vzorku nebyl prokázán.

Abstract:

This diploma thesis compares genotoxic properties of hydrogen peroxide and potassium permanganate by using the Comet Assay. The applications of hydrogen peroxide and potassium permanganate to the cells in the medium and cell lines immersed in the agarose gel were compared. The samples were applied for the HaCaT and A549 cell lines. DNA percentage in the comet tails was evaluated by using the CASP and LUCIA Comet Assay software. These factors were tested if they influence DNA disruption rate in the comet tails. The influence of the applied concentration of hydrogen peroxide, the method of the application, the type of the cell line and the software choice were discovered when using hydrogen peroxide. When applying potassium permanganate, the influence of the potassium permanganate concentration and the software choice were discovered only when sample was applied to the cells in the medium. Differences in DNA disruption between the cell lines were detected. The influence of the type of the sample application was not proved.
