Stability of biological material for examination of lymphocyte subpopulations by flow cytometry

Abstract:

Flow cytometry is a modern analytical method. It is used for the analysis of suspensions of particles or cells that are carried by a stream of carrier fluid to a light source. The light source in this case is a laser or a group of lasers which, together with an optical collection system, produce an optical signal. This is converted into an electronic signal by a photo-multiplier tube. The method has a wide spectrum of applications in the field of medicine and beyond. Specifically, in this bachelor thesis, flow cytometry had applications in the analysis of the stability of lymphocyte subpopulations. In the practical part, 20 patient whole blood samples were analyzed, 10 with physiological and 10 with pathological lymphocyte populations. All samples were measured over a period of 5 days. On the second day, after the analysis was completed, each sample was divided equally into 2 tubes. One tube continued to be stored at laboratory temperature and the other in a refrigerator at 2-8 °C. Each sample was analyzed in a doublet and the resulting value was obtained by averaging. These values were presented as box plots and then statistically processed using regression analysis. From the results of the statistical analysis, we found that there were no statistically significant changes in stability over time for the samples stored in the refrigerator. In contrast, for unrefrigerated samples, we found 7 cases where stability changes were statistically significant, but never for samples with increased lymphocyte populations. Based on the evaluation of the clinical significance of changes in the determined results over time, the stability of the biological material for the examination of lymphocyte subpopulations was set at 24 hours after collection when stored at laboratory temperature. Storage of samples without lymphocytosis at 2-8 °C can extend the stability of the material to 48 hours. The results of the work were compared with the literature, which focused on the stability of lymphocyte subpopulations for the purpose of their immunophenotyping. In this comparison, there were agreement or partially similar conclusions.

Abstract:

Prietoková cytometria predstavuje modernú analytickú metódu. Slúži na analýzu suspenzie častíc alebo buniek, ktoré sú unášané prúdom nosnej kvapaliny k svetelnému zdroju. Svetelným zdrojom je v tomto prípade laser alebo skupina laserov, ktoré spolu so zberným optickým systémom vytvárajú optický signál. Ten je následné prevedený na elektronický signál prostredníctvom fotonásobiča. Metóda má široké spektrum uplatnení v oblasti medicíny ale aj mimo nej. Konkrétne v tejto bakalárskej práci mala prietoková cytometria uplatnenie pri analýze stability lymfocytárnych subpopulácií. V praktickej časti bolo analyzovaných 10 pacientskych vzoriek plnej krvi, pričom 10 vzoriek bolo s fyziologickým a 10 s patologickým zastúpením lymfocytov. Všetky vzorky boli merané po dobu 5 dní. Druhý deň, po ukončení analýzy, bola každá vzorka rovnomerne rozdelená do 2 skúmaviek. Jedna skúmavka bola naďalej skladovaná pri laboratórnej teplote a druha v chladničke pri teplote 2-8 °C. Každá vzorka bola analyzovaná v doublete a výslednú hodnotu sme získali spriemerovaním. Tieto hodnoty boli prezentované vo forme krabicových grafov a následne štatisticky spracované pomocou regresnej analýzy. Z výsledkov štatistickej analýzy sme zistili, že u vzoriek skladovaných v chladničke nedošlo ku štatisticky významným zmenám stability v čase. Naopak u nechladených vzoriek sme zistili 7 prípadov, kedy zmeny stability boli štatisticky významné, avšak nikdy u vzoriek so zvýšeným zastúpením lymfocytov. Na základe zhodnotenia klinickej významnosti zmien stanovených výsledkov v čase bola stanovená stabilita biologického materiálu pre vyšetrenie subpopulácií lymfocytov na 24 hodín od odberu pri skladovaní pri laboratórnej teplote. Skladovanie vzoriek bez lymfocytózy pri teplote 2-8 °C môže predĺžiť stabilitu materiálu na 48 hodín. Výsledky práce boli porovnávané s literatúrou, ktorá sa zaoberala stabilitou subpopulácií lymfocytov za účelom ich imunofenotypizácie. Pri tomto porovnávaní došlo ku zhode alebo čiastočne podobným záverom.
