Bioactivity of polyene antibiotics produced by hemolytic streptomycetes

Abstract:

Tato bakalářská práce se zabývá bakteriemi rodu Streptomyces, které běžně obývající půdu, ale jsou považovány za kosmopolitní a řada kmenů je schopna obývat některé extrémní biotopy. V ojedinělých případech se jedná o bakterie patogenní, v přírodě však vytváří spíše symbiotické vztahy. Streptomycety jsou považovány za nejdůležitější zdroj bioaktivních látek. Produkují totiž řadu klinicky využívaných antibiotik, dále látky s antifungálními, protinádorovými, kancerostatickými, protizánětlivými, imunosupresivními a cytotoxickými vlastnostmi. Produkty sekundárního metabolismu těchto grampozitivních bakterií jsou i polyenová antibiotika - látky s výraznou antifungální aktivitou a také s hemolytickou aktivitou pozorovatelnou při kultivaci kultur na krevním agaru. Cílem experimentální části této práce bylo především ověření antifungálních vlastností půdních izolátů streptomycet vůči vybraným zástupcům vláknitých hub (plísní), dále byly zkoumány antibakteriální vlastnosti i vzájemné interakce streptomycet. Vyhodnocení zmíněných vlastností probíhalo na základě ko-kultivačních experimentů. Tato metoda imituje přirozeně se vyskytující podmínky života streptomycet, které jsou součástí často se měnících biotopů. Interakce mezi jednotlivými mikroorganismy byly vyhodnoceny na základě morfologických změn kultur a inhibičních zón. V případě interakcí mezi kmeny streptomycet byly interakce posuzovány na základě hemolytických zón. Dosažené výsledky naznačují rozmanitou biologickou aktivitu cytolytických metabolitů polyenové či nepolyenové povahy, produkovaných půdními izoláty streptomycet. Metabolity výrazně inhibovaly růst nejběžnějšího lidského fungálního patogenu - Candida albicans. Také plísně rodu Paecilomyces byly vůči sloučeninám citlivé. Plísně rodu Aspergillus a Fusarium byly až na některé výjimky rezistentní. Ko-kultivace streptomycet s bakteriálními patogeny neprokázaly žádné výrazné antibakteriální aktivity vůči grampozitivním (Staphylococcus aureus, Enterococcus faecalis) nebo gramnegativním (Escherichia coli) bakteriím. Některé interakce kmenů streptomycet s mikroby se zdály být specifické, což může naznačovat produkci doposud neznámých metabolitů.

Abstract:

This Bachelor thesis deals with important bacteria of the genus Streptomyces which are commonly soil dwellers but are considered cosmopolitan, and some strains can inhabit some extreme biotopes. In rare cases, they are pathogenic bacteria, because in nature they create rather symbiotic relationships. Streptomyces are considered to be the most important source of bioactive compounds. They produce several clinically used antibiotics, including substances with antifungal, antitumor, cancerostatic, anti-inflammatory, immunosuppressive, and cytotoxic properties. Other products of the secondary metabolism of these Gram-positive bacteria are polyene antibiotics - substances with significant antifungal activity, as well as hemolytic activity observable when cultivating cultures on blood agar. The experimental part of this work aimed to check the antifungal properties of the soil strains of Streptomyces against selected representatives of filamentous fungi (molds). The antibacterial properties and mutual interactions between Streptomyces were also investigated. The evaluation of the mentioned properties took place based on co-culture experiments. This method imitates the naturally occurring living conditions of Streptomyces, which are part of frequently changing biotopes. Interactions between individual microorganisms were evaluated based on morphological changes and growth inhibition zones. In the case of the interactions between Streptomyces strains, interactions were evaluated based on zones of hemolytic activity. The achieved results indicate the diverse biological activity of polyene or non-polyene cytolytic metabolites produced by soil-derived Streptomyces. The metabolites significantly inhibited the growth of the most common human fungal pathogen - Candida albicans. The fungi of the genus Paecilomyces were also sensitive to these compounds. The fungi Aspergillus spp. and Fusarium spp. were, with some exception, resistant. The co-cultures of Streptomyces with bacterial pathogens showed no significant antibacterial activities against Gram-positive (Staphylococcus aureus, Enterococcus faecalis) or Gram-negative (Escherichia coli) bacteria. Some interactions of Streptomyces strains with microbes appeared to be specific, which may indicate the production of yet unknown metabolites.
