Vliv genetických modifikací membránového kardiolipinu u <I>Streptococcus zooepidemicus</I> na produkci kyseliny hyaluronové

Effect of genetic modifications of membrane cardiolipin in <I>Streptococcus zooepidemicus</I> on hyaluronic acid production

Abstract:

Průmyslová produkce kyseliny hyaluronové (HA) fermentací bakterie <i>Streptococcus zooepidemicus</i> čelí výzvě v podobě přeměny části buněčné populace na nemukoidní fenotyp, což vede ke snížené produkci HA. Jedním z faktorů, který může ovlivnit tuto přeměnu, je interakce mezi enzymem hyaluronan syntázou a membránovým fosfolipidem kardiolipinem. Tato diplomová práce se v teoretické části věnuje enzymu hyaluronan syntázy, vlivu lipidů na jeho aktivitu, kardiolipinu a fenotypové přeměně u bakterií. V rámci experimentální části byly vytvořeny dva kmeny <i>S. zooepidemicus</i> – s delecí v genu kardiolipin syntázy a s dodatečnou kopií genů kardiolipin syntázy a fosfatidylglycerolfosfát syntázy, pro mukoidní i nemukoidní fenotyp. Následně byl sledován vliv těchto genetických modifikací na obsah kardiolipinu v membránách. V rámci diplomové práce bylo zjištěno, že narušení syntézy HA u nemukoidního fenotypu není způsobeno nedostatkem kardiolipinu v membránách. Nemukoidní fenotyp vykazoval dokonce statisticky významně vyšší obsah kardiolipinu oproti mukoidnímu fenotypu u wild-type kmene a dodatečná kopie genů způsobila statisticky významné snížení množství kardiolipinu u nemukoidního fenotypu. U mukoidního fenotypu <i>S. zooepidemicus</i> byl kromě obsahu kardiolipinu sledován vliv na výtěžek a molekulovou hmotnost HA. Výsledky ukázaly, že kmen s dodatečnou kopií genů nevykazoval zvýšenou produkci kardiolipinu a zároveň nebyl pozorován vliv na výtěžek a molekulovou hmotnost HA. U kmene s delecí v genu kardiolipin syntázy se podařilo narušit syntézu kardiolipinu. I přes nepřítomnost tohoto fosfolipidu v membránách byl pozorován mukoidní fenotyp kolonií značící zachování produkce HA. Ačkoliv se kvůli velkým rozptylům dat nepodařilo prokázat vliv na molekulovou hmotnost, bylo zjištěno snížení výtěžku HA. Důležitým výsledkem diplomové práce je, že kardiolipin není esenciální pro produkci HA a jeho role může být u <i>S. zooepidemicus</i> nahrazena jinými fosfolipidy.

Abstract:

Industrial production of hyaluronic acid (HA) by fermentation of <i>Streptococcus zooepidemicus</i> faces the challenge of converting part of the cell population to a non-mucoid phenotype, leading to reduced HA production. One factor that may influence this conversion is the interaction between the enzyme hyaluronan synthase and the membrane phospholipid cardiolipin. In this thesis, the theoretical part deals with the enzyme hyaluronan synthase, the influence of lipids on its activity, cardiolipin and phenotypic switching in bacteria. In the experimental part, two strains of <i>S. zooepidemicus</i> were created - with a deletion in the cardiolipin synthase gene and with an additional copy of the cardiolipin synthase and phosphatidylglycerol phosphate synthase genes for both mucoid and non-mucoid phenotypes. Subsequently, the effect of these genetic modifications on the cardiolipin content in membranes was investigated. It was found that the disrupted HA synthesis in the non-mucoid phenotype is not due to a lack of cardiolipin in the membranes. The non-mucoid phenotype even showed a statistically significantly higher cardiolipin content compared to the mucoid phenotype in the wild-type strain, and additional genes copy caused a statistically significant reduction in cardiolipin content in the non-mucoid phenotype. In the mucoid phenotype of <i>S. zooepidemicus</i>, the effect on the yield and molecular weight of HA was investigated in addition to cardiolipin content. The results showed that the strain with the additional genes copy did not show increased cardiolipin production, while no effect on yield and molecular weight of HA was observed. The strain with a deletion in the cardiolipin synthase gene has disrupted cardiolipin synthesis. Despite the absence of cardiolipin in the membranes, a mucoid phenotype of the colonies was observed, indicating preservation of HA production. Although an effect on molecular weight could not be demonstrated due to the large scatter in the data, a decrease in HA yield was observed. A significant result of this thesis is that cardiolipin is not essential for HA production, and other phospholipids in <i>S. zooepidemicus</i> may replace its role.
