Molecular biodiversity inventory of selected fish species from families Cyprinidae and Umbridae.

Abstract:

Doktorská práce se zabývá inventarizací molekulární diverzity vybraných taxonů dvou čeledí Cyprinidae a Umbridae na národní i evropské úrovni. Spojovacími prvky jsou rozsáhlá molekulárně-genetická analýza zahrnující analýzy jaderných i mitochondriálních markerů a vývoj mikrosatelitových lokusů. Práce je tematicky složená ze čtyř volně navazujících částí. Inventarizace biodiverzity ichtyofauny ČR byla založena na sekvenační analýze mitochondriálního a jaderného markeru. Projekt byl zpracován pod záštitou iniciativy Barcode of Life metodou „DNA barcoding“ a významně rozšířil mezinárodní databázi BoLD. Poskytl důležité informace pro monitorovací systém NATURA 2000 i novou druhovou kolekci pro Národní muzeum ČR. Fylogeografická studie vybraných druhů rodu Alburnoides odhalila 17 evropsko-asijských linií. Byl nalezen vhodný linijně-identifikační klíč složený z analýzy mitochondriálního markeru a indelové diagnostiky dvou jaderných genů. Byla potvrzena druhová podhodnocenost tohoto rodu. Byla podpořena validita 11 morfologicky uznaných druhů a čtyři nové linie byly předloženy jako „species-in-waiting“. Pro nominotypický druh Alburnoides bipunctatus s. stricto byl vymezen nový distribuční areál. Pro populační studie rodu Alburnoides byla připravena nová knihovna polymorfních mikrosatelitových lokusů. Použitelnost těchto lokusů byla testována při cross-species amplifikacích zástupců rodu Alburnoides a dalších čtyř ohrožených cyprinidů. Při rozlišování vybraných druhů ouklejky se některé testované mikrosatelity projevily jako diagnostické. Konstrukcí knihovny nových STR lokusů pro blatňáka tmavého jsme se významně podíleli na doplnění malého již publikovaného mikrosatelitového setu. Tyto mikrosatelitové lokusy jsou použitelné pro budoucí repatriační a populační studie na českém a evropském území.

Abstract:

The doctoral thesis is focused on molecular biodiversity inventory of selected fish species from two families, Cyprinidae and Umbridae. Linking elements are extensive molecular-genetic analysis concluding testing of nuclear and mitochondrial markers and testing of microsatellite loci. This thesis is thematically composed of four freely follow-up parts. The biodiversity inventory of ichthyofauna of the Czech Republic was based on analyses of the mitochondrial and nuclear markers. The project was processed under the auspices of the initiative Barcode of Life by method “DNA barcoding” and widened the database BoLD. This project also contributed to the monitoring system NATURA 2000 and for National Museum. Phylogeographical study of selected species of the genus Alburnoides revealed 17 Eurasian lineages. A good lineage-identification key composed of mitochondrial marker and indel diagnostics of nuclear genes was determined. Species underestimation within this genus was confirmed. The validity of 11 morphological valid species of genus Alburnoides was supported. Four lineages are considered as “species-in-waiting”. A new distribution area was determined for nominotypical species Alburnoides bipunctatus s. stricto. The new microsatellite library of polymorphic microsatellite loci for population studies was constructed. Usability of the loci was tested in the cross-species amplifications with individuals of genus Alburnoides and another four endangered cyprinids. Chosen species of spirlin are recognisable by using some microsatellites which were proved as diagnostic. We constructed the library of STR loci from the genome of European mudminnow and enriched the published small set of microsatellite loci. These microsatellites will be possible to use for repatriation and population studies in the Czech Republic and also widely in Europe.
