Analysis of p62/SQSTM1 expression in relation with exosomal secretion

Abstract:

Autofagie je autodegradativní proces, který se podílí na udržování homeostázy a vyrovnává zdroje energie v buňce recyklací poškozených a nadbytečných organel, proteinů a dalších makromolekul. Jedním ze substrátů podléhajících autofagické degradaci je protein p62. Jelikož se tento protein v důsledku inhibice autofagie hromadí a při indukci autofagie je jeho hladina nízká, může být použit jako marker pro studium autofagické aktivity. Proces autofagie je úzce spjat s exozomální sekrecí, jelikož oba tyto mechanismy sdílí některé své komponenty. Míra exozomální sekrece se typicky zvyšuje při defektech procesu autofagie. Cílem této práce je sledování závislosti exprese proteinu p62 nádorovou buněčnou linií FaDu, odvozenou z karcinomu oblasti hlavy a krku, na expresi exozomálních tetraspaninů za různých podmínek a úrovní buněčného stresu. Výsledkem je zjištění, které z vybraných látek indukují, respektive inhibují autofagii a jaký z toho plyne další efekt na nádorové buňky.

Abstract:

Autophagy is defined as a degradative process which participates in the maintenance of homeostasis and balances the energy sources in the cell by recycling the damaged or unwanted organelles, proteins, and other macromolecules. Protein p62 is one of the substrates that are subject to autophagic degradation, and is used as an indicator of the level of autophagy. The autophagy process is closely related to exosomal secretion, as they both share certain functional components. The level of exosomal secretion typically increases when defects in autophagy occur. In this thesis we track the dependence of protein p62 expression by the cell line FaDu derived from head and neck cell carcinoma on the expression of exosomal tetraspanins under different conditions and at different cellular stress levels. As the result of this thesis we find which of the selected substances act as atophagy inductors and inhibitors, respectively and what other effects they have in cancer cells
