Interakce plazmidů virulence a plazmidů přenášejících antibiotické rezistence u salmonel

Interaction of Salmonella virulence plasmids with plasmids encoding resistances to antibiotics

Abstract:

Antibiotic - resistant bacteria represent a serious public health issue of the 21st century. Antibiotic resistance is mostly encoded extra chromosomally within genetic elements. Resistance plasmids (R - plasmids) carry resistance to antibiotics and other growth inhibitors. Different R - plasmids in one cell can recombine with other plasmids. This is confirmed by existence of virulence plasmids that can recombine with the plasmids of resistance and generate plasmids with entirely new combinations of genes. Resistance plasmid p9134ΔTet, recombined with virulence plasmid pSLT could represent a relevant example. These plasmids can spread between enterobacteria and thus, give the emergence of multidrug-resistant pathogens with increased virulence. This thesis deals with the mutual interaction of resistance plasmids in Salmonella virulence plasmids. The thesis is supported with empirical evidence and results from conjugation experiments which used donor strain Salmonella serovar Typhimurium, that carried resistance plasmid termed p9134. This plasmid contained the resistance genes for resistance to ampicillin, chloramphenicol, streptomycin and tetracycline. Serovars S. Dublin 2229, S. Enteritidis 147, S. Choleraesuis G52, S. Gallinarum 140/5 and S. Typhimurium F98 were used as a recipient strains. In the first step, I gained complete transkonjugants of serovars S. Dublin, S. Enteritidis, S. Choleraesuis and S. Gallinarum, which contained the resistance plasmid. I checked each serovar of transkonjugants with serological test and I verified individual resistance genes as well as the presence of virulence plasmids with PCR method. In the second step, I observed recombination (failure resistance) in the conjugation of these transkonjugants to plasmid - free S. Typhimurium F98. Although I have seen the transfer of plasmid p9134 into recipient strain S. Typhimurium F98, I was unable to capture any failure resistance, which is hidden behind already mentioned resistance plasmid recombination with virulence plasmid. I compared the similarity of virulence plasmids serovars S. Dublin, S. Enteritidis, S. Choleraesuis and S. Gallinarum with plasmid p9134. By comparing, I found out that resistance plasmid p9134 has the greatest potential to recombine with virulence plasmid serovar S. Gallinarum pSGV.

Abstract:

V poslednom období dochádza k nárastu patogénov rezistentných na antibiotiká. Tento vážny problém 21. storočia sa týka verejného zdravia na celom svete. Antibiotická rezistencia býva najčastejšie kódovaná extrachromozómalne na genetických elementoch. Plazmidy rezistencie (R - plazmidy) nesú rezistenciu na antibiotiká ako aj na iné rastové inhibítory. Rozličné R - plazmidy v jednej bunke môžu rekombinovať s inými plazmidmi. Príkladom sú plazmidy virulencie, ktoré dokážu rekombinovať s plazmidmi rezistencie za vzniku plazmidov s novou kombináciou génov. Príkladom je plazmid rezistencie p9134ΔTet, u ktorého došlo k rekombinácii s plazmidom virulencie pSLT. Tieto plazmidy sa dokážu šíriť medzi enterobaktériami a dávajú tak vznik multirezistentným patogénom so zvýšenou virulenciou.Táto diplomová práca sa zaoberá vzájomnou interakciou plazmidov rezistencie s plazmidmi virulencie u salmonel. Pre konjugačné experimenty bol vybraný donorový kmeň sérovaru Salmonella Typhimurium, ktorý niesol plazmid rezistencie označovaný ako p9134. Tento plazmid rezistencie obsahoval gény pre rezistenciu na ampicilín, chloramfenikol, streptomycín a tetracyklín. Ako recipientné kmene boli použité sérovary S. Dublin 2229, S. Enteritidis 147, S. Choleraesuis G52, S. Gallinarum 140/5 a S. Typhimurium F98. V prvom kroku som získala kompletných transkonjugantov sérovarov S. Dublin, S. Enteritidis, S. Choleraesuis a S. Gallinarum, ktoré obsahovali tento plazmid rezistencie. Serotypizáciou som si overila sérovar jednotlivých transkonjugantov a metódou PCR som si overila jednotlivé gény rezistencie a prítomnosť plazmidov virulencie. V druhom kroku som pozorovala rekombináciu (výpadok rezistencie) pri konjugácii týchto transkonjugantov do bezplazmidového kmeňa S. Typhimurium F98. Síce som zaznamenala prenos plazmidu p9134 do recipientného kmeňa S. Typhimurium F98, ale nepodarilo sa mi zachytiť žiaden výpadok rezistencie, za ktorou je skrytá už spomínaná rekombinácia plazmidu rezistencie s plazmidom virulencie. Porovnala som podobnosť jednotlivých plazmidov virulencie sérovarov S. Dublin, S. Enteritidis, S. Choleraesuis a S. Gallinarum s plazmidom p9134. Z porovnania som zistila, že najväčší potenciál rekombinovať s plazmidom rezistencie p9134 má plazmid virulencie sérovaru S. Gallinarum pSGV.
