Regulation of interactions and activity of oncoproteins MDM2 and MDMX

Abstract:

Nádorový supresor p53 hraje klíčovou roli v regulaci buněčné odpovědi na nejrůznější formy buněčného stresu. Za normálních podmínek je hladina aktivního p53 v buňkách udržována na nízké úrovni především díky přítomnosti onkoproteinů Mdm2 a MdmX. Na druhou stranu, nadměrná exprese onkogenů Mdm2 a MdmX bývá příčinou inaktivace p53 u mnoha typů nádorů. Zatímco zvýšená exprese Mdm2 vede k degradaci p53 prostřednictvím proteasomu 26S, protein MdmX sám o sobě pouze inhibuje transkripční aktivitu p53. MdmX však může interagovat s Mdm2 a v tom případě se aktivně podílí i na degradaci p53. Cílem disertační práce bylo objasnit mechanismy regulující aktivitu komplexu Mdm2-MdmX, zejména analyzovat rozdíly mezi RING doménami proteinů Mdm2 a MdmX a blíže specifikovat funkci centrální kyselé domény Mdm2 v procesu degradace p53.

Abstract:

Tumor suppressor p53 has a key role in the regulation of cellular responses to various types of stress. Its function is kept under tight control in unstressed cells primarily by interactions with oncoproteins Mdm2 and MdmX. On the other hand, overexpression of Mdm2 and MdmX is responsible for p53 inactivation in a significant proportion of tumors. While overexpression of Mdm2 leads to p53 degradation in 26S proteasome, the main function of MdmX alone is a direct inhibition of p53 transcriptional activity. However, MdmX is able to bind Mdm2 and in this way directly contributes to p53 degradation. The aim of this thesis was to shed light on mechanisms regulating activity of the Mdm2-MdmX complex, especially to determinate critical differences in the RING domains of Mdm2 and MdmX and to clarify the role of Mdm2 central acidic domain in p53 degradation.
