DNA barcoding of amphibians in a case study of the batrachofauna of the Dzanga-Sangha Forest Reserve, Central African Republic

Abstract:

DNA barcoding je poměrně jednoduchý a praktický molekulárně-genetický nástroj k základnímu odhalování druhové identity, případně diverzity vyšších taxonů. Praktické využití má zejména u morfologicky kryptických či málo známých taxonů nebo těžce určitelných ontogenetických stadií. Tato práce poukazuje na základní problematiku a vývoj DNA barcodingu u obojživelníků. Pomocí zmíněné metody je zde přiblížena dosud málo prozkoumaná druhová diverzita batrachofauny rezervace Dzanga-Sangha ve Středoafrické republice. V této studii bylo ze 106 vzorků porovnaných v databázi GenBank, zjištěno celkem 8 čeledí, 15 rodů 34 druhů, což činí přibližně 35% celkové předpokládané diverzity obojživelníků daného regionu severozápadní části Konžské pánve. Výsledná data poskytují lepší představu o druhové diverzitě obojživelníků rezervace Dzanga-Sangha, čehož může být využito pro lepší posouzení zranitelnosti a stanovení priorit ochrany druhů i celého ekosystému. Dílčí výsledky budou využity také v taxonomickém zhodnocení vybraných druhových komplexů.

Abstract:

DNA barcoding is relatively simple and practical molecular-genetic method originally developed for species identifications but commonly utilized in many fields of research, especially in the biodiversity science. The method has its main practical application in identifications of morphologically cryptic or little known taxa, or ontogenetic stages of difficult species identification. This study reviews methodology, utility and new progress of the DNA barcoding in amphibian research. This method is applied to study little-known diversity of the amphibian fauna of the Dzanga-Sangha Forest Reserve, located in rainforests of the southwestern Central African Republic. Altogether, 106 samples were analysed and compared with the database GenBank. The studied material covered 8 families, 15 genera and 34 species, which represented approximately 35% of expected amphibian diversity in the area of the northwestern Congo Basin. These results are the first information on the diversity of the amphibian fauna of the Dzanga-Sangha Forest Reserve, which is expected to provide additional insights into understanding of the overall biological richness of this unique ecosystem and will help its conservation. Results will be also utilized in future taxonomic studies of selected species complexes.
