Characterisation of newly derived medulloblastoma cell lines with special attention to the possibilities of induced differentiation

Abstract:

Meduloblastom je velmi heterogenní onemocnění s odlišnostmi mezi jednotlivými nádory. V současné době se vědci snaží najít nejlepší způsob, jak identifikovat jednotlivé podskupiny meduloblastomu (Wnt, Sonic Hedgehod a skupina C a D), které by umožnily rychlé stanovení typu nádoru a nasazení správné léčby. K této kategorizaci je nejčastěji využíváno expresního profilování, nicméně byla též publikována studie popisující rozdělení do těchto podskupin dle přítomnosti čtyř různých markerů – GAB1, YAP1, filamin A a β-catenin. Diplomová práce je zaměřena na charakterizaci meduloblastomových linií, a to jak referenčních, tak linií derivovaných z nádorové tkáně pacientů FN Brno. U těchto linií byla prokázána přítomnost markerů, které potvrzují neurální původ buněk. Jednalo se o vimentin, synaptofyzin, neuron-specifickou enolázu, nestin a tři druhy neurofilament, a dále i markery pro stanovení podskupin meduloblastomu. V rámci práce byla analyzována exprese markerů YAP1, GAB1, filamin A a β-catenin nepřímou imunofluorescencí, metodou western blotting a pomocí RT-PCR. Dále byl sledován vliv all-trans retinové kyseliny (ATRA) na expresi těchto markerů na úrovni mRNA. Všechny analyzované linie byly pozitivní na přítomnost cytoplazmatického filaminu A, β-cateninu, cytoplazmatického i jaderného YAP1. Rozdíly byly detekovány pouze u proteinu GAB1. Vliv ATRA na expresi genů β-catenin, filamin A, YAP1 a GAB1 nebyl prokázán. Z výsledků dále vyplývá, že fenotypové rozdíly mezi meduloblastomovými liniemi neodpovídají stanoveným podskupinám primárních nádorů a metoda klasifikace na základě přítomnosti těchto markerů je v případě meduloblastomových buněčných linií v podmínkách in vitro nevhodná.

Abstract:

Medulloblastoma is very heterogeneous disease because individual tumors are very different from each other. Currently, scientists try to find the best way to identify subgroups of medulloblastoma (Wnt, Sonic Hedgehog, group 3 and group 4). This stratification could be used for quick determination of medulloblastoma and for choice of best treatment strategy. This stratification is usually based on the expression profiling. However, a new study suggests four different markers (GAB1, YAP1, filamin A a β-catenin) to stratify medulloblastoma subgroups. The diploma thesis is focused on the characterization of medulloblastoma cell lines –both reference cell lines and cell lines derived from medulloblastoma tissues taken from patients in the University Hospital Brno. In this study, vimentin, synaptophysin, nestin, neuron-specific enolase and three type of neurofilaments as markers of neuronal origin, and also markers for identification medulloblastoma subgroups, i. e. YAP1, GAB1, filamin A and β-catenin were analyzed by indirect immunofluorescence, western blotting and RT-PCR. The influence of all-trans retinoic acid (ATRA) on the expression of these markers at mRNA level was also evaluated. All cell lines were positive for cytoplasmic filamin A and β-catenin, cytoplasmic and nuclear YAP1. Differences were observed only in the expression of GAB1 protein. No influence of ATRA on gene expression of these markers was observed. These results showed that differences in phenotype among these cell lines did not correspond to these of primary tumors and this classification is not suitable in medulloblastoma cell lines under in vitro conditions.
