Epigenetic processes in mammals

Abstract:

Epigenetika je obor zabývající se změnami genové exprese, které nejsou způsobeny přímou změnou v sekvenci nukleotidů daných genů. Zaměřením tohoto oboru je tedy popis široké škály chemických modifikací informačních makromolekul (DNA, RNA a proteinů, zejména histonů) a zároveň jejich ovlivnění vnějšími faktory prostředí. Jak název práce napovídá, jedná se zejména o problematiku tohoto oboru promítnutou do řízení genové exprese u savců. Úvodní kapitola stručně shrnuje myšlenkové proudy a teorie celkového kontextu evolučních teorií a epigenetiky, doplněno o významné objevy na poli molekulární biologie, týkajících se zejména epigenetické dědičnosti. Je tak vytvořeno chronologické upořádání informací o historii tohoto oboru. Kapitoly první poloviny bakalářské práce popisují epigenetické mechanismy na molekulární úrovni. Účelem těchto textů je co nejpodrobnější popsání pomyslných základních kamenů, na jejichž interakcích stojí vlastní specifické projevy fenotypu. Druhá část práce je již striktně zaměřena na epigentické procesy charakteristické pro taxon savců. Tyto kapitoly jsou sepsány podle určitého vzoru, popisujícího historii objevu daného jevu, jeho obecnou charakteristiku, molekulární mechanismus a vlastní projev. Závěrečná kapitola je poté věnována krátkému popisu chorob souvisejících s nesprávnou funkcí epigenetických mechanismů. V textu jsou častokrát zmiňovány analogie epigenetických procesů jiných taxonů a čtenář si tak může vytvořit alespoň letmý pohled na epigenetiku z evolučního hlediska. Text je rovněž doplněn obrázky a schématy pro lepší pochopení dané problematiky.

Abstract:

Epigenetics is a science dealing with changes in gene expression that are not due to a direct change in the nucleotide sequence of the genes. The focus of this field is a description of a wide range of chemical modification of information macromolecules (DNA, RNA and proteins, mainly histones) as well as environmental factors have influence on them. As the job title suggests, the thesis is focused to the control of gene expression in mammals. The first chapter summarizes the evolutionary theory and epigenetics, complemented by important discoveries in the field of molecular biology, in particular regarding epigenetic inheritance. This creates a chronological arrangement of information about the history of this industry. First chapters of thesis describe epigenetic mechanisms at the molecular base. The purpose of these texts is to describe the basic mechanisms of the regulation of DNA expression. The second part of the thesis is strictly focused on epigentics processes of characteristic taxon - mammals. These chapters describe the discovery of the science, its general characteristics, molecular mechanism and self expression. The final chapter is dedicated to a short description of the diseases associated with the malfunction of epigenetic mechanisms. In text there are often mentioned analogy epigenetic processes of other taxons. You can find there also pictures and diagrams to better understand this processes.
