In vivo toxicity evaluation of heteroborans using model organism D. rerio

Abstract:

Některé sloučeniny boru, např. heteroborany (COSANy), jsou člověkem uměle vytvořené látky. Své uplatnění mohou najít ve farmakologii a medicíně. Z toho vyplývá nutnost testovat toxicitu těchto látek před jejich případným uvedením do klinické praxe. K zjištění negativních vlivů nových látek se uplatňují mnohé in vitro i in vivo testy. Pro tuto predikci se využívá různých modelových organismů. Diplomová práce se zaměřuje na charakteristiku nejvyužívanějších modelových organismů pro testování toxicity. Vzhledem k zaměření experimentální části práce je větší důraz kladen na FET test prováděný na embryích Danio rerio. Ten slouží jako jeden z prvotních nástrojů k určení toxicity látek, ať už nových nebo běžně využívaných. Pomocí FET testu je v této práci stanovena a srovnána toxicita 4 heteroboranů, jakožto nových a dosud nezkoumaných látek.

Abstract:

Boron compounds, in particular heteroborans (COSANs), are man-made substances with potential application in pharmacology and medicine. This brings the need to test the toxicity of these substances before introducing them to clinical use. Many in vitro and in vivo tests can be used to determine the negative effects of novel substances. Different model organisms could be used for toxicity evaluation. The diploma thesis focuses on the characteristics of the most used model organisms for toxicity testing. Due to the experimental part of the work, more emphasis is given on the FET test performed on Danio rerio embryos. It is used as one of the primary tools for determining the toxicity of substances, either novel or commonly used. Using this test, the toxicity of 4 heteroboranes was evaluated and compared in this work.
