The use of stem cells in the treatment of cancer

Abstract:

Bakalářská práce je zaměřena na použití mezenchymálních kmenových buněk jako buněčného přenašeče pro léčbu nádorových onemocnění, a to jak teoreticky, tak s použitím experimentu. Teoretická část se na úvod věnuje nádorovým onemocněním, konkrétně příčinám jejich vzniku, prevalenci a klasifikaci. Rakovina je stále jednou z nejčastějších příčin úmrtí po celém světě. Důvody, proč k maligním změnám dochází, bývají komplexní povahy. Následující oddíl teoretické části je pak zaměřen na standardní péči o pacienty s rakovinou, a na nové buněčné terapeutické přístupy, které jsou stále na počátku zrodu, ale mohou se stát dalším milníkem v léčbě rakoviny. S odkazem k různým buněčným terapeutickým přístupům popisuje bakalářská práce použití a možnosti modifikace mezenchymálních kmenových buněk, a to jako nosiče protinádorového agens pro léčbu rakoviny. Díky jejich unikátnímu nádorovému tropismu a velmi snadné izolaci z lidského těla patří mezi nejslibnější kandidáty pro budoucí buněčnou terapii nádorových onemocnění. Co se týká části praktické, mezenchymální kmenové buňky byly modifikovány k expresi protinádorového agens TRAIL. Jejich terapeutický účinek byl pak hodnocen analýzou jejich cytotoxického účinku proti buňkám nádorových linií MDA-MB-231, RPMI 8226 a U2OS. Získané výsledky pak ukazují, že mezenchymální kmenové buňky, modifikované k expresi TRAIL, mohou vykazovat významný cytotoxický účinek vůči různým testovaným nádorovým liniím.

Abstract:

The bachelor thesis consists of the use of mesenchymal stem cells as a cell vehicle to treat cancer, using a theoretical and an experimental approach. The theoretical part deals first with cancer illnesses, their causes of origin, prevalence, and classification. Cancer is still one of the most common causes of death worldwide. The reasons for malignant changes tend to be of a complex nature. The following section, corresponding to the theoretical part, focuses on the cancer standard of care and the new cellular therapeutic approaches, which are still fledgling but can become the next pillar in cancer treatment. Regarding the different cellular therapeutic approaches, the work describes the use and engineering of the mesenchymal stem cells as a cellular vehicle to treat cancer. Because of their unique tumor tropism and easy isolation from the human body, they are among the most promising candidates for the future cellular treatment of cancer. Concerning the practical section, the mesenchymal cells were modified for the expression of the antitumor agent TRAIL. Their therapeutic effect was assessed by analyzing their cytotoxic effect against the tumor cell lines MDA-MB-231, RPMI 8226, and U2OS. The results obtained show that mesenchymal stem cells, after modified for TRAIL expression, can exert a significant cytotoxic effect against the different tumor cell lines tested.
