The dietery induction of hyperglycaemia at the female diabetic rat ZDF and the study of the influence hyperglycaemia to wound healing

Abstract:

Diabetes mellitus 2. typu je nejčastější metabolické onemocnění, které vzniká v důsledku relativního nedostatku inzulinu. Buňky nejsou schopné přijímat glukózu, dochází k hyperglykémii. Právě následkem dlouhodobé hyperglykémie dochází ke vzniku nejrůznějších zdravotních komplikací, k nejzávaţnějším patří porucha procesu hojení ran. Cílem práce bylo zjistit, jak diety s různým obsahem tuku ovlivňují vývoj diabetu u samic potkanů Zucker Diabetic Fatty (ZDF). Potkani ZDF mají mutaci v genu leptinového receptoru, coţ způsobuje jeho defekt a leptin se tak nemůţe navázat na centrum sytosti v mozku. Potkan trpí pocity hladu a při dostatku potravy se u něj vyvíjí obezita a následně i diabetes mellitus 2. typu. Samice ZDF potkanů byly rozděleny do dvou skupin na diabetické (genotyp fa/fa) a kontrolní. Pro sledování závislosti vývoje diabetes mellitus 2. typu byly kaţdé skupině po dobu 4. - 18. týdnů podávány čtyři typy speciálních diet s rozdílným obsahem tuku: H1 (24,6 % tuku), H2 (33,2 %), Z (C13004, 25,6 %) a St1 (3,4 %). Během experimentu byly u těchto zvířat sledovány a popsány změny v hmotnostech a glykémiích. Dále bylo metodou ELISA stanoveno, jak se vyvíjejí plazmatické koncentrace inzulinu v závislosti na dietě u kontrolních a diabetických zvířat. Pokus byl ukončen ve věku zvířat 16. - 20. týdnů vytvořením excizní kruhové rány a sledováním jejího hojení po dobu deseti dní. Desátý den byla z rány odebrána granulační tkáň. Z ní byla vyizolována RNA a metodou rt RT-PCR byla provedena analýza exprese genu pro MMP-3. Naše výsledky ukázaly, ţe pro vyvolání a udrţení hyperglykémie je nevhodnější dieta H1. Pro vyvolání stabilní, lehce zvýšené glykémie (10 mmol/l) a hyperinzulinemie je vhodná dieta Z. Dieta H2 je velmi nevhodná. Hojení ran a exprese MMP-3 se v rámci jednotlivých diet významně nelišily.

Abstract:

Diabetes mellitus type 2 is the most common metabolic disease that arises due to a relative lack of insulin. The cells are not able to accept glucose which results in hyperglycemia. Long-term hyperglycemia leads to serious complications when a failure of wound healing is one of the most serious of them. The aim of the thesis was to determine how the diets, with different contain of fat, affect the development of diabetes in female Zucker Diabetic Fatty (ZDF) rats. ZDF rats have mutation in gene coding leptin receptor that causes its defect and leptin cannot bind to the satiety center in the brain. The rat is hungry and if it gets sufficiency of food, obesity and subsequently diabetes mellitus type 2 develops. Female ZDF rats were assigned to one of two groups, the diabetic (genotype fa/fa) or control one. For monitoring of dependence of development the diabetes mellitus type 2, there were introduced four types of special diets with a different content of fat to all groups within a period of 4 - 18 weeks. The following diets were used: H1 (24,6 % fat), H2 (33,2 %), Z (C13004, 25,6 %) a St1 (3,4 %). Within the experiment, the changes in weights and glycaemia in diabetics of the animals were observed and described. Moreover, the plasmatic concentration of insulin in dependence on diet of healthy and diabetic animals was evaluated by using ELISA method. The experiment was terminated when the animals were 16 - 20 weeks old and an excision circle wound was formed. Healing of this wound was observing for ten days. The granulation tissue, from which RNA was isolated, was taken from the healing wound on the tenth day. The analysis of MMP-3 gene expression was performed by using rt RT-PCR method. Our results indicated that for inducing and retaining hyperglycaemia, the most suitable diet is the H1 one. For inducing a stable, slightly increased glycaemia (10 mmol/l) and hyperinsulinemia, the appropriate diet is the Z one. The diet H2 is very unsuitable. The differences in wounds healing and expression MMP-3 within particular diets were not significant.
