Abstract:

V předkládané práci byly identifikovány tři základní patologie, u kterých selhává farmakologická léčba a není dořešen optimální způsob dopravení bioaktivních látek do cílové tkáně. Těmito patologiemi jsou ischemie myokardu, degradace kloubní chrupavky, progredující kožní rána (zejména diabetická). Proto teoretická i praktická část disertace obsahuje vždy tři stěžejní kapitoly se speciálním zaměřením na určitou problematiku. Byly navrženy originální metody modifikace exosomů a generování liposomů bohatých na specifická miRNA. Společným jmenovatelem testovaných metod jsou klíčové komponenty, které je možno označit jako: liposomy, exosomy, nanodelivery systémy a sono-elektrická stimulace. Metoda pro léčbu chrupavky a kožní patologie vykazuje velmi dobré kurativní výsledky.

Abstract:

In the presented work, three basic pathologies were identified, in which pharmacological treatment fails and the optimal method of delivering bioactive substances to the target tissue has not been resolved. These pathologies are ischemia of the myocardium, degradation of articular cartilage, progressive skin wound (especially diabetic). Therefore, the theoretical and practical part of the dissertation always contains three key chapters with a special focus on a certain 3 pathologies. Original methods for modifying exosomes and generating liposomes rich in specific miRNAs have been proposed. The common denominator of the tested methods are key components, which can be called as: liposomes, exosomes, nanodelivery systems and sono-electrical stimulation. The final method for the treatment of cartilage and skin pathology shows very good curative results.
