DNA polymorphism analysis in selected flax collection (Linum usitatissimum L.)

Abstract:

Tato diplomová práce je složena ze 2 částí, teoretické a praktické. V teoretické části je vytvořen stručný přehled o lnu setém, jeho původu, využití, pěstování a šlechtění. Kromě samotného šlechtění lnu jsou zde vypracovány i kapitoly o šlechtění obec-ně, včetně historie. Další část se zabývá genetickou diverzitou a uchováváním genových zdrojů v genových bankách. V závěru literární části je také zmíněno využití molekulárních markerů. V praktické části jsou uvedeny informace o laboratorní analýze 32 odrůd lnu setého. Vybranou metodou byla molekulární analýza s iPBS markery. Díky ní bylo získáno, s použitím 25 primerů, celkem 284 produktů. Míra polymorfismu dosahovala 95,77 %. Z fylogenetického stromu lze v závěru vyčíst, jak jsou vybrané odrůdy mezi sebou odlišné, a které mají společný původ.

Abstract:

This diploma thesis consists 2 parts, theoretical and practical. The theoretical part provides a brief overview of flaxseed, its origin, use, cultivation and breeding. Besides to flax breeding itself, there are also chapters about breeding in general, including its history. Next part deals with genetic diversity and conservation of gene resources in gene banks. The use of molecular markers is also mentioned at the end of the litera-ture part. Practical part contains informations about laboratory analyses of 32 varieties of flaxseed. The chosen method was molecular analysis with iPBS markers. Thanks to it, a total of 284 products were discovered by using 25 primers. The polymorphism rate was 95.77 %. In conclusion, the phylogenetic tree shows how selected varieties are different from each other and which of them have a common origin.
