Deregulation of Cell Proliferation by Polycyclic Aromatic Hydrocarbons with High Molecular Weight

Abstract:

Polycyklické aromatické uhlovodíky (PAU) patří mezi významné kontaminanty životního prostředí, které vznikají nedokonalým spalováním organických látek. V rámci této diplomové práce byly studovány některé účinky skupiny PAU o molekulové váze 278 na buněčné úrovni, protože některé předchozí studie naznačily jejich možné karcinogenní účinky. PAU o molekulové váze 278 (dibenzanthraceny a benzochryseny) jsou, podobně jako polychlorované dibenzo-p-dioxiny nebo polychlorované bifenyly, ligandy Ah receptoru. V důsledku jeho aktivace dochází nejen k indukci enzymů podílejících se na metabolické aktivaci PAU, ale i k dalším efektům na buněčné úrovni. AhR má i celou řadu funkcí, které souvisí s regulací buněčné proliferace, apoptózy či diferenciace, což by mohlo hrát roli v nádorově promočních účincích AhR ligandů. AhR ligandy způsobují v hepatocytech a celé řadě nádorových buněčných linií zástavu buněčného cyklu v G1 fázi, zároveň ale mohou indukovat buněčnou proliferaci v jaterních progenitorových buňkách potkana. V rámci této diplomové práce bylo zjištěno, že studované látky způsobovaly proliferaci konfluentních jaterních progenitorových buněk potkana WB-F344. Účinné byly především dibenzanthraceny. Mezi benzochryseny je pozorovatelný významný nárůst proliferace u benzo[b]chrysenu a benzo[c]chrysenu. Naopak u benzo[g]chrysenu byly pozorovány především účinky vedoucí k programované buněčné smrti (apoptóze) a velmi výrazné akumulaci buněk v S fázi buněčného cyklu. Na základě literárních údajů i těchto výsledků lze předpokládat, že tyto jeho účinky souvisely s jeho genotoxickými vlastnostmi. Předchozí studie ukázaly, že proliferační účinky PAU souvisí s aktivací AhR, která vede k nárůstu exprese cyklinu A, který se významným způsobem podílí jak na regulaci S fáze, tak obecně na regulaci buněčné proliferace. Všechny studované PAU indukovaly zvýšený počet buněk v S fázi v závislosti na koncentraci. Výrazný nárůst byl pozorován především po působení dibenz[a,h]anthracenu a benzo[g]chrysenu. Bylo prokázáno, že tento nárůst koresponduje s výrazně zvýšenou expresí cyklinu A a byla tak potvrzena jeho role v účincích studovaných PAU. Bylo také zjištěno, že obě tyto látky, v závislosti na koncentraci, indukují expresi CYP1A1. Tento enzym je jedním z cílových genů AhR a bylo tak tedy prokázáno, že mechanismus působení této skupiny PAU se shoduje s již publikovanými daty o jiných skupinách PAU. Závěrem lze konstatovat, že PAU s molekulovou váhou 278 mají schopnost narušovat regulaci buněčného cyklu v modelové linii jaterních progenitorových buněk, a tento jev souvisí s jejich schopností aktivovat Ah receptor a indukovat expresi cyklinu A. Narušení kontroly buněčné proliferace by mohlo přispívat ke karcinogenním účinkům této skupiny PAU.

Abstract:

Polycyclic aromatic hydrocarbons (PAHs) belong among important environmental contaminants, which are produced by incomplete combustion of organic matter. In the present diploma thesis, some effects of a group of PAHs with molecular weight 278 at the cellular level were investigated, because previous studies implied their potential carcinogenic effects. PAHs with molecular weight 278 (dibenzanthracenes and benzochrysenes) are, similar to polychlorinated dibenzo-p-dioxins or polychlorinated biphenyls, ligandy Ah receptor. Its activation leads not only to induction of enzymes participating in metabolic activation of PAHs, but also to other effects at the cellular level. AhR has a number of functions related to regulation of cell proliferation, apoptosis and differentiation, which might play a role in tumor promoting effects of AhR ligands. AhR ligands induce G1 block in hepatocytes or in a number of tumor cell lines; on the other hand, they can induce cell proliferation in rat liver progenitor cells. Within this work, it was found that the compounds under study induced proliferation of confluent rat liver progenitor WB-F344 cells. Dibenzanthracenes were particularly efficient. Among benzochrysenes, a significant increase in cell proliferation was observed in case of benzo[b]chrysene and benzo[c]chrysene. In contrast, benzo[g]chrysene induced effects leading to programmed cell death (apoptosis) and a highly pronounced accumulation of cells in the S phase of cell cycle. Based both on the literary data and the present findings, it could be assumed that these effects were due to its genotoxic properties. Previous studies suggested that the proliferative effects of PAHs are due to activation of the AhR, which leads to increased cyclin A expression, which plays an important role both in S phase regulation and generally also in regulation of cell proliferation. All studied PAHs induced accumulation of cell in S phase in a concentration-dependent manner. A significant increase was observed especially after treatment with dibenz[a,h]anthracene and benzo[g]chrysene. It was found that this increase corresponds with an increased expression of cyclin A, thus confirming its role in the effects of PAHs under study. It was also found that both compounds induce expression of CYP1A1 in a concentration-dependent manner. This enzyme is one of the AhR target genes, therefore implying that the mode of action of these PAHs is comparable to published data on other groups of PAHs. In conclusion, it could be stated that PAHs with molecular weight 278 might deregulate cell cycle in the model cell line of rat liver progenitor cells, and this effect is related to their ability to activate Ah receptor and induce cyclin A expression. Disruption of cell proliferation control might contribute to carcinogenic activity of this particular group of PAHs.
