Dynamics of epidermal UV-shielding and protective function of carotenoids in selected species of higher plants

Abstract:

Tato diplomová práce se zabývá UV-stíněním a fyziologickým stavem rostlin ječmene jarního pěstovaných při vysoké a nízké ozářenosti. Také zkoumá, jak reaguje ječmen jarní na přisvětlování UV-A zářením. Pro nahlédnutí do problematiky byla zpracována stručná teoretická část. Ta popisuje UV-stínění, ochrannou funkci karotenoidů, ječmen jarní a použité metody. Byly provedeny dva experimenty. Při prvním byly porovnávány rostliny ječmene jarního divokého (WT) a mutantního typu chlorina f2 (CLO) pěstované při dvou ozářenostech (1000 a 50 ?mol m-2s-1). Při druhém experimentu byly porovnávány rostliny přisvětlované UV-A zářením a kontrolní rostliny pěstované při dvou ozářenostech (1000 a 50 ?mol m-2s-1). U prvního experimentu jsme zjistili, že míra UV-stínění je u WT rostoucího při vysoké ozářenosti (HI) vyšší než u rostlin rostoucích při ozářenosti nízké (LI). Gradient UV-stínění podél listu je výraznější u WT než u CLO a u rostlin pěstovaných při LI ve srovnání s HI rostlinami. Rostliny divokého typu pěstované při HI vykazují 6.den pokles parametru FV/FM a vzestup SV0 související se stárnutím listů. Není patrný trend zvyšování tepelné disipace u rostlin HI po dobu vývoje, to svědčí o optimálním využití absorbovaného světla uvnitř PS II. Z druhého experimentu jsme usoudili, že větší vliv na fyziologický stav rostliny má FAR než přisvětlování UV-A radiací. Rostliny rostoucí při HI jsou ve větším stresu, snaží se aktivovat obranné mechanismy a stárnou rychleji. UV-A radiace může u HI snížit obsah chlorofylů a karotenoidů. Relativní obsah luteinu a pigmentů xantofylového cyklu (VAZ) se naopak při UV-A u HI zvyšuje. UV-A nemá vliv na zvyšování UV-stínění. UV-A záření při HI snižuje maximální fotochemickou účinnost PS II a zvyšuje míru nezářivé disipace, pří LI tento vliv nemá.

Abstract:

This thesis deals with the UV-shielding and physiological status of barley plants grown at high and low irradiance. It also examines how spring barley responds to the exposure of UV-A radiation. For insight into the issue brief theoretical part was prepared. It describes the UVshielding, protective function of carotenoids, spring barley and methods used. Two experiments were carried out. In the first one wild-type barley (WT) and its mutant chlorina f2 (CLO) grown at two irradiances (1000 and 50 ?mol m -2s-1) were compared. In a second experiment, UV-A radiation-exposed and control plants grown at two irradiances (1000 and 50 ?mol m -2s-1) were compared. In the first experiment, we found that the rate of UV-shielding of the WT grown at high irradiance (HI) was higher than in plants grown at low irradiance (LI). UVshielding gradient along the leaf is more pronounced in WT than in CLO and plants grown at LI compared with HI plants. WT grown at HI in the 6th day showed a decrease of parameter FV/FM and rise of SV0 associated with aging of leaves. There was no apparent trend of increasing capacity of thermal dissipation in LI plants throughout the development, it shows on the optimal use of absorbed light within the PS II. From the second experiment, we deduced that the FAR has a greater influence on the physiological status of plants than UV-A radiation. Plants growing in HI are more under stress trying to activate defense mechanisms and age faster. UV-A radiation at HI can reduce the content of chlorophylls and carotenoids. The relative content of pigments lutein and xanthophyll cycle (VAZ) is contrary to UV-A in HI increases. UV-A has not the effect of increasing UV-shielding. UV-A radiation in HI plants reduces the maximal photochemical efficiency of PS II and increase the rate of thermal dissipation and in LI plants UV-A radiation has not this effect.
