Determination of selected keto acids in dried blood spot using high-performance liquid chromatography with fluorescence detection

Abstract:

Stanovení aminokyselin a od nich odvozených 2-oxokyselin má klíčovou úlohu v diagnostice vrozených metabolických poruch u dětí v rámci novorozeneckého screeningu. V biochemické laboratoři s klasickým vybavením se pro stanovení 2-oxokyselin využívá jako "zlatý standard" metoda vysokoúčinné kapalinové chromatografie s fluorescenční detekcí, při které se 2-oxokyseliny derivatizují ortho-fenylendiaminem. Jedná se o poměrně citlivou metodu, avšak ortho-fenylendiamin je vysoce karcinogenní látka. Tato práce se zabývá stanovením vybraných 2-oxokyselin v suché kapce krve metodou HPLC s fluorescenční detekcí, s cílem nahradit ortho-fenylendiamin za jiné nekarcinogenní derivatizační činidlo a zachovat, či zvýšit citlivost dané metody. Pro derivatizaci vybraných 2-oxokyselin byly zvoleny tři derivatizační činidla, 1,2-diamino-4,5-dimethoxybenzen, 1,2-diamino-4,5-methylendioxybenzen a dansylhydrazin. Při použití dansylhydrazinu je nevýhodou to, že tato látka fluoreskuje stejně jako deriváty 2-oxokyselin. U metody využívající 1,2-diamino-4,5-methylendioxybenzen jako derivatizační činidlo se nám nepodařilo ve vzorku suché kapky krve dostatečně separovat deriváty vybraných 2-oxokyselin. Byla zavedena metoda HPLC s fluorescenční detekcí pro stanovení vybraných 2-oxokyselin ve vzorcích suché kapky krve. Jako derivatizační činidlo byl použit 1,2-diamino-4,5-dimethoxybenzen, přičemž vzniklé deriváty mnohem silněji fluoreskovaly v porovnání s metodou využívající pro derivatizaci 2-oxokyselin ortho-fenylendiamin. Deriváty 2-oxokyselin byly separovány v systému obrácených fází (analytická kolona LiChroCart 125 x 4 mm, Purospher Star RP-18e, 5 um, opatřená ochranou kolonou LiChroCart 4 x 4 mm, Purospher Star RP-18e, 5 um) gradientovou elucí. Mobilní fází A byla směs acetonitrilu a deionizované vody (25:75; v/v) a mobilní fází B byl 100% acetonitril. Přesnost v sérii pro stanovení vybraných 2-oxokyselin ve vzorcích suché kapky krve byla vyhovující, variační koeficienty se pohybovaly pod 5 %. Kalibrační křivky byly lineární v rozsahu testovaných koncentrací, s výjimkou 2-oxobutyrátu, kde byl průběh z nejasných důvodů kvadratický. Orientační hladiny vybraných 2-oxokyselin u 14 dobrovolných dárců jsou porovnatelné s literárními údaji. Fenylpyruvát byl detegován jen u některých dárců krve.

Abstract:

The determination of amino acids and keto acids play a key role in the diagnosis of congenital metabolic diseases, which is part of the new-born screening. The HPLC method with fluorescence detection is "golden standard", which is used for a determination of keto acids in biochemical laboratories with a basic equipment. The HPLC method uses ortho-phenylenediamine for the derivatization of keto acids. It is relatively sensitive method, but ortho-phenylenediamine is a highly carcinogenic substance. This thesis deals with the determination of keto acids in dried blood spots by an HPLC with fluorescence detection and replacing ortho-phenylenediamine with another non-carcinogenic derivatizing agent to maintain or increase the sensitivity of the method. Three derivatizing agents were chosen for a derivatization of keto acids, 1,2-diamino-4,5-dimethoxybenzene, 1,2-diamino-4,5-methylenedioxybenzene and dansylhydrazine. The major disadvantage of dansylhydrazine is that it emits fluorescence as much as derivatives of keto acids. We didn't succeed in a sufficient separation of keto acids in dried blood spots after the derivatization with 1,2-diamino-4,5-methylenedioxybenzene. The HPLC method with fluorescence detection of keto acids in dried blood spots was established. 1,2-Diamino-4,5-dimethoxybenzene was used as the derivatizing agent. The derivatives of keto acids emit fluorescence much more than the derivatives after the derivatization with ortho-phenylenediamine. The derivatives of keto acids were separated by reversed-phase HPLC with gradient elution (an analytical column LiChroCart 125 x 4 mm, Purospher Star RP-18e, 5 um, connected with guard column LiChroCart 4 x 4 mm, Purospher Star RP-18e, 5 um). The mobile phase A was a mixture of acetonitrile and deionized water (25:75; v/v) and mobile phase B was 100% acetonitrile. The intra-assay precision of presented HPLC method was satisfactory, the coefficients of variation were under 5 %. The calibration curves were linear over the tested range for keto acids except 2-ketobutyrate, calibration curve was quadratic, the reason is unclear. The orientation levels of selected keto acid in dried blood spots obtained from 14 volunteers are comparable to literature data. We detected phenyl pyruvate only in some blood donors.
