External Factors Influencing the Production of Biogenic Amines in Selected Grampositive Bacteria

Abstract:

V bakalářské práci je přiblížen vliv faktorů, které ovlivňují produkci biogenních aminů u vybraných grampozitivních bakterií. Konkrétně byla sledována kinetika produkce biogenních aminů u kmene Lactobacillus brevis RIBM 2-69 při 0; 3 a 6% (v/v) koncentraci etanolu v růstovém prostředí. Lactobacillus brevis je řazen mezi možné kontaminanty piva, které jsou navíc schopné zvýšené produkce biogenních aminů, čímž negativně ovlivňují kvalitu tohoto alkoholického nápoje, případně i zdraví konzumenta. Vznik biogenních aminů (tyraminu, fenyletylaminu, putrescinu, kadaverinu) byl po kultivaci sledován v supernantantu pomocí kapalinové chromatografie na reverzních fázích s předkolonovou derivatizací danzylchloridem a UV detekcí. U testovaného mikroorganizmu bylo uskutečněno sledování kinetiky produkce tyraminu. Právě tyramin byl produkován v hojné míře, na rozdíl od ostatních BA, jejichž koncentrace nedosáhla ani 2 mg/l.

Abstract:

In the bachelor thesis the factors affecting the production the biogenic amines in selected grampositive bacteria is described. The kinetics of biogenic amine production by Lactobacillus brevis RIBM 2-69 at 0, 3 and 6 % (v/v) concentration of ethanol in the growth medium was observed. Lactobacillus brevis is considered to be one of the most possible beer contaminants. These bacteria are able to produce higher levels of biogenic amines which negatively affect the quality of alcoholic drinks and subsequently the consumer's health. The formation of biogenic amines (tyramine, phenylethylamine, putrescine, cadaverine) was monitored after the cultivation in the supernatants by reverse phase liquid chromatography with pre-column derivatization by dansylchloride and UV detection. The observation of kinetics of tyramine production was realized. Only tyramine was produced by the current tested bacterial culture. The tyramine was formed in abundant measure in comparison with concentrations of other biogenic amines which were bellow 2 mg/l.
