Analyses of Steroids in the Feather and Plasma of Birds

Abstract:

Základní funkce mnoha steroidních hormonů včetně testosteronu a kortikosteronu zastávají mezi obratlovci stejnou roli. U ptáků jsou jejich koncentrace úzce spojeny s morfologickými, behaviorálními a dalšími fyziologickými rysy včetně reprodukce, metabolismu, imunitního systému a fyzické zdatnosti. Vliv hormonů na výše zmíněné vlastnosti a potenciální vazba mezi hormony jsou předmětem zkoumání. Hlavním cílem této práce je rozšířit naše znalosti o steroidních hormonech v ptačím peří a krevní plasmě ptáků, a to konkrétně ze dvou pohledů. Zaprvé, zhodnotit analýzu steroidů, vyvinout metody pro jejich detekci a kvantifikaci i při velmi nízkých hodnotách koncentrací a validovat analytické parametry pro stanovení steroidů v biotických matricích. Zadruhé, zhodnotit vztah mezi měřenými hladinami steroidních hormonů a vybranými charakteristikami organismu. K dosažení těchto cílů jsou v dizertační práci předloženy tři studie. První studie byla zaměřena na testování podmínek přípravy vzorku peří. Druhá studie hodnotila, jak jsou koncentrace steroidních hormonů v peří spojeny s pohlavím, věkem a mírou růstu ocasních per malých stěhovavých ptáků. Třetí studie sledovala vliv matricového efektu na stanovení steroidů v různých zdrojích krevní plazmy. Prezentované výsledky poskytují nástroj pro spolehlivé studium změn koncentrací vybraných steroidních hormonů v biotě, chování steroidů při zpracování vzorků a identifikaci steroidů. Vhodné použití analytické metody navíc zvyšuje kvalitu shromažďovaných dat a následně jejich vyhodnocení. Výsledky této práce tak mohou přispět k lepšímu pochopení fyziologických mechanismů organismů.

Abstract:

The main functions of many steroid hormones, including testosterone and corticosterone, have the same roles between vertebrates. In birds, their concentrations are closely connected with many morphological, behavioural, and other physiological traits, including reproduction, metabolism, the immune system, and fitness. The direction of the effect of these hormones on the abovementioned traits, and the potential feedback between hormones are the subject under investigation. The main goal of this thesis is to contribute to our wider knowledge about steroid hormones in the feathers of birds and blood plasma, specifically through two objectives: (1) to evaluate steroid analysis, i.e. to develop methods for their detection and quantification even at very low concentration and carry out validation of the analytical parameters for determination of steroids in biotic matrices; and (2) to evaluate the association between measured steroid hormone levels and selected characteristics of the organisms. To fulfil these objectives, three studies are presented. The first study focused on the test condition of feather sample preparation. The second study evaluated how feather steroid concentrations are associated with sex, age, and tail feather growth rates in a small migratory passerine. The third study focused on the matrix effect on determination of steroids in different sources of blood plasma. The presented results provide a tool to reliably study the changes in the level of concentration of selected steroid hormones in the biota, as well as the behaviour of steroids during sample processing, and also to identify steroids. Moreover, appropriate use of the analytical method improves the quality of collected data and data applications. The results of this thesis can thus contribute to a better understanding of the physiological mechanisms of organisms.
