Preparation of recombinant 17ß-HSD10 protein and in vitro study of its inhibition

Abstract:

V rámci této disertační práce byla vypracována rešerše shrnující současné poznatky o enzymu 17β-hydroxysteroid dehydrogenasa typu 10 (17β-HSD10). V této práci byla podrobně popsána jak fyziologická, tak patologická role tohoto enzymu v buněčném metabolismu a jeho zapojení v patologii Alzheimerovy nemoci a maligních onemocnění. Byly popsány dosud publikované inhibitory tohoto enzymu. Na základě dostupné literatury bylo potvrzeno, že tento enzym je vhodným cílem pro vývoj cílených inhibitorů.Vzhledem k velkému množství sloučenin s potenciálním inhibičním účinkem na tento enzym, které byly k dispozici na pracovišti, byla hledána vhodná metoda pro jejich testování. V rámci experimentální práce byla vypracována metodika pro rekombinantní produkci enzymu 17β-HSD10 v bakteriálních buňkách, v nichž byl enzym úspěšně exprimován a purifikován. Následně byla optimalizována metoda pro stanovení aktivity a byly stanoveny kinetické parametry enzymu, které byly porovnatelné s daty ve dříve publikovaných pracích.Vyvinutou metodou byly testovány potenciální inhibitory enzymu z interní databáze sloučenin. Na základě získaných výsledků byly selektovány nejlepší sloučeniny a byla určena hodnota jejich IC50 a kinetika inhibice enzymu. Mezi nejsilnější inhibitory patřily látky se strukturou 6-substituovaného benzothiazolu spojeného močovinovým spojovacím řetězcem se 3-Cl, 4-OH substituovaným fenylem. Tyto sloučeniny měly hodnotu IC50 v nízké mikromolární koncentraci a byl u nich určen akompetitivní typ inhibice. Jejich interakce s proteinem 17β-HSD10 byla dále potvrzena metodou diferenční skenovací fluorimetrie.

Abstract:

As a part of this dissertation thesis, a literature review summarizing current knowledge about the enzyme 17β-hydroxysteroid dehydrogenase type 10 (17β-HSD10) was prepared. In this paper, both the physiological and pathological role of this enzyme in cellular metabolism and its involvement in the pathology of Alzheimer's disease and cancer were determined. 17β-HSD10 inhibitors published to date were described. Based on the available literature, it was confirmed that this enzyme is a suitable target for the development of targeted low-molecular inhibitors.Due to a large amount of compounds with potential inhibitory ability towards 17β-HSD10 that were available on site, a methodology for their evaluation was researched. As a part of experimental work, a methodology for recombinant protein production of 17β-HSD10 in E. coli was developed. The enzyme was successfully expressed and purified. Subsequently, the assay for determination of its enzymatic activity was optimized and the kinetic parameters were found. The obtained data were comparable with previously published literature.The developed method was used for testing of potential inhibitors from the internal database. Based on the results, the best compounds were selected and their IC50 values and kinetics parameters were determined. The most potent inhibitors were those with the structure of a 6-substituted benzothiazole linked by urea linker with 3-Cl, 4-OH substituted phenyl ring. These compounds have the IC50 values in low micromolar concentration and their uncompetitive type of inhibition was determined. Their interaction with 17β-HSD10 was further confirmed by differential scanning fluorimetry.
