Characterization of nucleoprotein complexes in plant order Asparagales

Abstract:

Telomery jsou specializované nukleoproteinové struktury na koncích eukaryotních chromozomů. U většiny vyšších eukaryot jsou tvořeny repetitivní dvouřetězcovou DNA. Jeden z telomerických řetězců obsahuje velké množství guaninů (G), tzv. G řetězec. Jeho 3´ konec tvoří jednořetězcový přesah. Ten slouží jako substrát pro telomerázu, reverzní transkriptázu, enzymu, který používá svoji RNA podjednotku jako templát pro syntézu telomerických sekvencí na koncích chromozomů. Další důležitou vlastností G-přesahu je jeho schopnost vnořit se do dvouřetězcové oblasti telomery, čímž chrání konce chromozomů. Nepřehlédnutelnou roli v těchto procesech hrají telomer-vazebné proteiny, především takové, které se váží na jednořetězcový přesah. Tato práce byla zaměřena na analýzu telomer-vazebných proteinů se specifitou k jednořetězcovému G přesahu rostlin řádu Asparagales. Tento řád je zajímavý z evolučního hlediska, protože u některých druhů této skupiny byly „typické“ rostlinné telomery (TTTAGGG)n plně nebo částečně nahrazeny lidským typem telomerových repetic (TTAGGG)n. V této práci jsme sledovali interakce mezi jadernými proteiny rostlin Muscari armeniacum (modřenec) a Scilla peruviuna (ladoňka) (čeleď Hyacinthaceae, řád Asparagales) a telomerickými oligodeoxynukleotidy rostlinného (TTTAGGG)6 – AttelG a lidského (TTAGGG)6 – HutlG typu. Pomocí retardační analýzy a SDS-PAGE jsme zjistili, že jaderné proteiny modřence tvoří jeden komplex s radioaktivně značeným oligodeoxynukleotidem AttelG a jeden se značeným oligodeoxynukleotidem HutlG. Mezi jadernými proteiny ladoňky a rostlinnou sondou (AttelG) se vytváří několik nukleoproteinových komplexů. Naproti tomu tyto proteiny a savčí typ oligodeoxynukleotidu (HutlG) tvoří pouze dva specifické komplexy. Pro ověření vazebné specifity těchto proteinů k jednořetězcovému G přesahu, jsme testovali jejich schopnost tvořit komplexy s komplementárními C řetězci a dvouřetězcovými sondami. Tato analýza potvrdila vazebnou specificitu ke G řetězci. Dalšími pokusy jsme prokázali, že některé z komplexů jsou stabilní i při vysokých teplotách. Kontrolní experiment s proteinázou potvrdil, že sledované komplexy jsou výsledkem DNA – proteinových interakcí. Vzhledem k tomu, že některé proteiny váží své cílové sekvence jako monomery, homomultimery či heteromultimery, studovali jsme také vazebné schopnosti proteinů separovaných podle jejich molekulové hmotnosti. Na základě tohoto experimentu můžeme předpokládat, že většina charakterizovaných proteinů se na telomery váže jako monomer nebo homomultimer. Naše výsledky ukazují, že se v jaderných extraktech obou rostlin nachází telomer vazebné proteiny se specifitou k jednořetězcovému G přesahu jak rostlinného tak savčího typu, ačkoliv silnější vazbu vykazují k typu původnímu, tedy rostlinnému.

Abstract:

Telomeres are specialized structures at the ends of eukaryotic chromosomes. In higher eukaryots they are formed by double-stranded DNA while their end is formed by 3’ overhang of a G-rich strand. This overhang serves as substrate for telomerase, reverse transcriptase, enzyme which uses an internal RNA template to synthesize telomeric repeat sequences onto chromosome ends. G overhang also protects chromosome ends due to its ability to nest into double-stranded telomeric region. Telomere binding proteins, especially telomere G-strand-binding ones play important roles in these processes. This work was focused to characterization of telomere binding proteins with specificity to G-rich strand from the plant order Asparagales. This order is interesting from evolutionary aspect, because in some species of this group “typical” plant telomeres (TTTAGGG)n have been partially or fully replaced by human-type repeats (TTAGGG)n. We studied interactions between nuclear proteins from Muscari armeniacum and Scilla peruviana (family Hyacinthaceae, order Asparagales) and telomeric oligodeoxynucleotides of plant (TTTAGGG)6 – AttelG, and human (TTAGGG)6 – HutlG type. By the retardation analysis and SDS-PAGE we found out, that nuclear proteins from Muscari formed one single complex with radioactively end-labelled oligodeoxynucletide AttelG and another with HutlG. A few protein complexes were detected between Scilla nuclear proteins and AttelG and two complexes were found with HutlG. To test binding specificity of studied proteins to G- rich strand, complementary C-strand and double-stranded telomeric oligodeoxynucleotides were used as probes in retardation analysis. This experiment confirmed specificity to G-strand. Moreover, we have found that some nucleoprotein complexes are quite heat-stable. Control experiment with proteinase confirmed, that all retarded bands are results of DNA-protein interactions. Since some DNA-binding proteins bind their target sequences as monomers or homo/heteromultimers, we tried to test binding activities of particular nuclear proteins fractionated according to their molecular weight. On the basis of this experiment we can suppose, that most of studied proteins bind their target sequence as monomers or homomultimers. In conclusion, there are telomere binding proteins with binding specificity to G-rich strand in both nuclear protein extracts. These proteins are able to bind both types of telomere sequences, while the plant type of sequence shows stronger binding.
