Determination of Dissociation Constants of the Drug Bedaquiline

Abstract:

V této diplomové práci jsem stanovovala smíšené disociační konstanty léčiva Bedaquilinu UV-VIS spektrofotometrickou titrací a potenciometrickou titrací při 15°C, 25°C, 37°C, 40°C a 50°C. Data byla vyhodnocena regresním programem REACTLAB, ESAB, SQUAD84 a faktorovou analýzou INDICES. Nelineární regresí smíšené disociační konstanty v závislosti na iontové síle byla vyčíslena termodynamická disociační konstanta pro všechny teploty a z naměřených disociačních konstant při 5 teplotách byla vyčíslena entropie a entalpie.

Abstract:

In this master thesis I determined the mixed dissociation constants of the Bedaquiline medicine by UV-VIS spectrophotometric titration and potentiometric titration at 15°C, 25°C, 37°C, 40°C and 50°C. Data were evaluated by regression program REACTLAB, ESAB, SQUAD84 and factor analysis INDICES. The thermodynamic dissociation constant for all temperatures was calculated by nonlinear regression of the mixed dissociation constant as a function of ionic strength, and the entropy and enthalpy were calculated from the measured dissociation constants at 5 temperatures.
