The interactome of telomerase RNA

Abstract:

Telomeráza, ribonukleoproteinový komplex, který hraje zásadní roli při udržování fyzických konců lineárních chromozomů (telomer) se skládá ze dvou hlavních komponent: telomerázové reverzní transkriptázy (TERT) a telomerázové RNA podjednotky (TR). Kromě těchto dvou hlavních částí je kompletní telomeráza tvořena také dalšími proteiny, které jsou nezbytnou součástí pro její správné fungování in vivo. Holoenzym telomerázy společně s dalšími proteiny, zabraňuje zkracovaní telomer, ke kterému dochází během každého buněčném dělení. Avšak složení těchto proteinových komplexůse může lišit napříč různými větvemi evolučního stromu. U Arabidopsis thaliana byla telomerázová RNA podjednotka (AtTR) objevena v naší laboratoři teprve nedávno, ale proteiny s ní asociované zůstávají stále předmětem výzkumu. Na základě toho jsme optimalizovali metodu založenou na interakci radioaktivně značených proteinů s různými druhy RNA fúzovaných s RNA aptamery. Tato metoda může sloužit jako účinný přístup nejen k ověření předpokládaných proteinů vážících se na AtTR, ale může být použita i k charakterizaci potenciálních vazebných partnerů libovolné RNA.

Abstract:

Telomerase is a ribonucleoprotein complex which is crucial for maintaining the length of telomeres, the physical ends of linear chromosomes. It is comprised of two pivotal components: telomerase reverse transcriptase (TERT) and telomerase RNA subunit (TR). Beyond these core components, additional proteins form part of the complete telomerase, essential for its proper in vivo function. Telomerase holoenzyme, together with additional proteins, prevents telomeric shortening that naturally occurs after each cell division. Remarkably, the composition of these protein complexes may vary across different branches of the evolutionary tree. In Arabidopsis thaliana, the telomerase RNA subunit (AtTR) was described in our laboratory only recently, but the proteins associated with this RNA subunit are still awaiting elucidation. To address this, we have optimised a method based on the interaction of radioactively labelled proteins with various kinds of RNAs fused with RNA aptamers. This method can serve as an efficient approach not only to validate putative AtTR binding proteins but can also be used to characterise potential binding partners of any RNA of interest.
