Vztah mezi modifikacemi DNA kancerogeny, mutageny a protinádorově účinnými látkami a jejich biologickou aktivitou

Relationship between DNA modifications by cancerogens, mutagens and antitumour agents and their biological activity

Abstract:

Cisplatina (cis-diammindichloridoplatnatý komplex) je významné protinádorové léčivo, které se využívá při léčbě nádorů varlat, vaječníků a močového měchýře. Navzdory velkému úspěchu cisplatiny, její využití v léčbě nádorových onemocnění je spojeno s několika omezeními, mezi které patří vedlejší účinky a relativně úzké spektrum cisplatinou léčitelných nádorů (mnohá nádorová onemocnění jsou k cisplatině rezistentní). S cílem překonat tyto nevýhody jsou syntetizovány nové koordinačních komplexy platiny či jiných kovů a ty pak dále studovány z hlediska jejich protinádorové účinnosti. V této práci byl studován mechanismus protinádorového působení nových platnatých komplexů s inhibitory cyklin-dependentních kináz (CDK), boheminem (2-(3-hydroxypropylamino)-6-benzylamino-9-isopropylpurin) (L1) a jeho dvěma deriváty (2-chloro-6-(benzylamino)-9-isopropylpurin) (L2) a (2-chloro-6-[(4methoxybenzyl)amino]-9-isopropylpurin) (L3), jako ligandy: cis-[Pt(L1)2Cl2] (C1), cis-[Pt(L2)2Cl2] (C2) a cis-[Pt(L3)2Cl2] (C3). Tyto bifunkční konjugáty byly syntetizovány se záměrem spojit v jedné molekule látky s odlišnými biologickými vlastnostmi, schopností vazby na DNA a s CDK inhibiční aktivitou. Výsledky předložené práce, alespoň částečně, odhalují buněčný metabolismus a vazebné vlastnosti na DNA těchto nových analogů cisplatiny. Z výše zmíněných komplexů se jako nejslibnější ukazuje komplex C1. Tento komplex C1 je schopný překonávat rezistenci k cisplatině v A2780cisR rezistentních buněčných liniích a navíc vykazuje nízkou toxicitu v nenádorových liniích. In vitro experimenty ukazují, že vazebný mód komplexu C1 na DNA je podobný módu cisplatiny. Identické typy aduktů komplexu C1 a cisplatiny však narušují konformaci DNA odlišně. Předkládaná práce se dále zabývá studiem vazebných vlastností na DNA "neplatinových" sloučenin, a to nově syntetizovaných komplexů ruthenia s arénovými ligandy a také antracyklinů – doxorubicinu a jeho mnohonásobně účinnějšího derivátu 2-pyrrolinodoxorubicinu. Pro tyto účely byla v práci využita metoda transkripčního mapování, která je metodou poskytující informace o vazbě těchto chemoterapeutik na DNA a také jejich vlivu na transkripční procesy.

Abstract:

Cisplatin (cis-diamminedichloridoplatinum(II)) is an effective antitumor metallodrug which is widely used in the treatment of testicular, ovarian and bladder carcinomas. Despite its success, cisplatin has several disadvantages, which include severe side effects and limited spectrum of curable tumors, as many tumors are resistant to cisplatin. To overcome these drawbacks various platinum and other metal complexes have been synthesized and their mode of antitumor action was further studied. The main part of the thesis deals with the study of the molecular mechanism of action of the new class of anticancer drugs, in which the cyclin-dependent kinases (CDKs) inhibitor bohemine 2-(3-hydroxypropylamino)-6-benzylamino-9-isopropylpurine (L1) and its analogues 2-chloro-6-benzylamino-9-isopropylpurine (L2) and 2-chloro-6-[(4methoxybenzyl)-amino]-9-isopropylpurine (L3) are coordinated to Pt(II) to form cisplatin derivatives: cis-[Pt(L1)2Cl2] (C1), cis-[Pt(L2)2Cl2] (C2) and cis-[Pt(L3)2Cl2] (C3). These bifunctional Pt-CDK inhibitory conjugates were designed with the intention to synthesize platinum complexes which would exhibit besides its DNA binding properties also a CDK inhibitory activity. Results of the present work revealed, at least partly, cellular processing and DNA binding properties of these new agents. Among above listed complexes, complex C1 showed to be the most promising one. Complex C1 is capable of circumventing the resistance to cisplatin in A2780cisR resistant cells and moreover this new analogue exhibits low toxicity in nontumorigenic cells. In vitro experiments suggested that several aspects of DNA binding mode of C1 and cisplatin were similar. However, it cannot be excluded that identical types of DNA adducts of C1 and cisplatin can distort DNA conformation differently and can be also differently processed by cellular components. Reported herein is also a study of DNA binding properties of non-platinum agents, new ruthenium complexes with arene ligands and anthracyclines – doxorubicin and its more potent analogue 2-pyrrolinodoxorubicin. The transcription mapping was employed to gain a better insight into DNA binding processes of these chemotherapeutics and their influence on transcription mechanisms.
