Isolation of ATB resistant bacteria and monitoring of the occurrence of resistome in river water

Abstract:

Diplomová práce se zabývá problematikou antibiotické rezistence a výskytem bakteriálního rezistomu ve vodním prostředí. Rešeršní část práce je zaměřena na charakteristiku antibiotik, jejich klasifikaci a mechanismus účinku. Další část práce se zabývá bakteriální rezistencí k antibiotikům, mechanismům bakteriální rezistence a podstatě vzniku rezistence. Pozornost je věnována také vstupu a kumulaci antibiotik ve vodním prostředí, rezistentním bakteriím a bakteriálnímu rezistomu ve vodním prostředí. V experimentální části jsou testovány vzorky vod z rybníku, do něhož odtéká vyčištěná odpadní voda z ČOV Lučina, z přehrady Žermanice a řeky Lučiny. U odebraných vzorků je stanoven počet všech kultivovatelných bakterií a počet kultivovatelných rezistentních bakterií k ampicilinu a tetracyklinu. Rezistentní izoláty jsou izolovány a charakterizovány stanovením minimální inhibiční koncentrace a minimální baktericidní koncentrace. U rezistentních izolátů jsou detekovány vybrané geny rezistence pomocí PCR metody. Následně je sledován transfer rezistentních bakterií a genů rezistence ve vodním prostředí.

Abstract:

The diploma thesis deals with the issue of antibiotic resistance and the occurrence of bacterial resistome in the aquatic environment. The research part of the work is focused on the characteristics of antibiotics, their classification and mechanism of action. The next part of the thesis deals with bacterial resistance to antibiotics, mechanisms of bacterial resistance and the nature of the of resistance formation. Attention is also paid to the application and accumulation of antibiotics in the aquatic environment, resistant bacteria and bacterial resistance in the aquatic environment. In the experimental part, water samples are tested from the pond into which the treated wastewater from the Lučina WWTP flows, from the Žermanice dam and the Lučina river. The number of all culturable bacteria and the number of culturable bacteria resistant to ampicillin and tetracycline are determined for the samples taken. Resistant isolates are isolated and characterized by determining the minimum inhibitory concentration and the minimum bactericidal concentration. In resistant isolates selected resistance genes are detected using the PCR method. Subsequently, the transfer of resistant bacteria and resistance genes in the aquatic environment is monitored.
