3D bioprinting for the study of immune system cells

Abstract:

Bakalářská práce je zaměřená na technologii 3D biotisku, která se hlavně využívá k výrobě 3D modelů tkání a orgánů. První část práce se věnuje popisu 3D biotisku. Zde je stručně vysvětlena historie, rozebrány a popsány 4 hlavní technologie 3D biotisku, dále jsou charakterizovány biomateriály, jejich vlastnosti a uvedeny konkrétní příklady biomateriálů, které jsou nepostradatelnou složkou pro 3D biotisk modelů tkání a orgánů. Práce se také zabývá imunitní odpovědí na implantaci a enkapsulaci biomateriálů a imunitní odpovědí na konkrétní biomateriály. Následující část práce je zaměřena na využití 3D biotisku. 3D biotisk nabízí širokou škálu aplikací, které zahrnují transplantace orgánů, testování a screening léků a výzkum rakoviny. Z těchto oblastí jsou uvedeny příklady modelů srdce, jater a rakovin vytvořených pomocí metody 3D biotisku. Dále jsou také uvedeny příklady využití 3D biotisku pro vytvoření nádorové mikroprostředí pro imunoterapii a využití 3D biotisku pro imunoterapii založenou na NK buňkách. Poslední část bakalářské práce se zabývá návrhem experimentu pro následující diplomovou práci, která bude mít název 3D biotisk pro studium patogeneze mléčné žlázy.

Abstract:

The bachelor's thesis is focused on 3D bioprinting technology, which is mainly used to produce 3D models of tissues and organs. The first part of the work is devoted to the description of 3D bioprinting. Here, the history is briefly explained, the 4 main technologies of 3D bioprinting are analyzed and described, biomaterials are further characterized, their properties and specific examples of biomaterials, which are an indispensable component for 3D bioprinting of tissue and organ models, are given. The work also deals with the immune response to the implantation and encapsulation of biomaterials and the immune response to specific biomaterials. The following part of the work is focused on the use of 3D bioprinting. 3D bioprinting offers a wide range of applications that include organ transplants, drug testing and screening, and cancer research. From these areas, examples of heart, liver and cancer models created using the 3D bioprinting method are given. Examples of the use of 3D bioprinting to create a tumor microenvironment for immunotherapy and the use of 3D bioprinting for immunotherapy based on NK cells are also presented. The last part of the bachelor's thesis deals with the proposal of the experiment for the following diploma thesis, which will be called 3D bioprinting for the study of the pathogenesis of the mammary gland.
