Study on metabolism of benzophenanthridine alkaloids

Abstract:

This work is focused on study of the reactions of phase I biotransformation of benzo[c]phenanthridine alkaloids by rat microsomal fraction. Firstly the behavior of this system is verified using sanguinarine and chelerythrine, whose metabolism has been described in literature. Then the metabolites of the minor alkaloids (chelirubine, chelilutine, sanguirubine, sanguilutine, and macarpine) are produced and identified by liquid chromatography coupled to high-resolution tandem mass spectrometry. Major types of reactions observed in this work are reduction to dihydroderivatives, O-demethylation, O-demethylenation, and hydroxylation.

Abstract:

Tato práce je zaměřena na studium reakcí první fáze biotransformace benzo[c]fenantridinových alkaloidů pomocí systému potkaní mikrosomální frakce. Nejdříve je ověřena funkčnost systému pomocí sanguinarinu a chelerytrinu, jejichž metabolismus je detailně popsán v literatuře. Dále jsou produkovány a identifikovány metabolity minoritních alkaloidů (chelirubinu, chelilutinu, sanguirubinu, sanguilutinu a makarpinu). Pro jejich identifikaci je využita kapalinová chromatografie spojená s vysokorozlišující tandemovou hmotnostní spektrometrií. Hlavními typy metabolických reakcí pozorovanými v této práci jsou redukce na dihydroderiváty, O-demethylace, O-demethylenace a hydroxylace.
