Mutagenesis of AFL1 lectin

Abstract:

The object of the study is the AFL1_RtoA mutant protein whose mutations are based on substitution of arginines in all six binding sites of AFL1 lectin by alanines. The mutations should result in total loss of the lectin ability to bind carbohydrates. This bachelor thesis builds on previous research focused on AFL1 lectin, which is considered as a virulence factor of the opportunistically-pathogenic fungus, Aspergillus fumigatus, and serves as a model system for the study of complex interactions. Experiments which were carried out in this work are a part of the project focused on a determination of specificity and affinity of each binding site of the AFL1 lectin individually via reverse mutations of one binding site of protein AFL1_RtoA at a time. Besides a literature review, the bachelor thesis contains a description of methods and results summarizing subcloning of the afl1_RtoA mutant gene into the pET28 and pET29 vectors and testing the expression and solubility of AFL1_RtoA in a bacterial expression system.

Abstract:

Předmětem studia je mutantní protein AFL1_RtoA , jehož mutace spočívají v záměně argininů ve všech šesti vazebných místech lektinu AFL1 za alaniny, se záměrem úplné ztráty schopnosti vázat sacharidy. Tato bakalářská práce navazuje na předchozí výzkum, zaměřený právě na lektin AFL1 , který je považován za virulentní faktor podmíněně-patogenní plísně Aspergillus fumigatus a slouží jako modelový systém pro studium komplexních interakcí. Experimenty, provedeny v této práci, jsou součástí projektu, zaměřeného na stanovení specifity a afinity každého vazebného místa lektinu AFL1 jednotlivě, a to prostřednictvím zpětných mutací vždy jednoho vazebného místa proteinu AFL1_RtoA zvlášť. Bakalářská práce obsahuje kromě literární rešerše i popis metod a výsledků, které sumarizují překlonování mutantního genu afl1_RtoA do vektorů pET28 a pET29 a testování exprese a rozpustnosti mutantního proteinu AFL1_RtoA v bakteriálním expresním systému.
