Functional characterization of PIWI-interacting RNA associated with glioblastoma multiforme pathology

Abstract:

Multiformní glioblastom (GBM) je nejčastějším primárním nádorem mozku s infaustní prognózou a mediánem přežití přibližně 12 až 15 měsíců od stanovení diagnózy. Biologická agresivita onemocnění a délka přežívání pacientů se však mezi jednotlivými případy mnohdy značně liší. Důvodem je vysoká heterogenita GBM jak na buněčné, tak molekulární úrovni. Bližší pochopení intracelulární signalizace a vzájemných vztahů mezi molekulami ovlivňujícími klíčové biologické vlastnosti GBM buněk by tak mohlo vést k objevení nejen nových terapeutických přístupů, ale i prognostických a prediktivních biomarkerů odpovědi na léčbu pacientů s tímto onemocněním. Moderními vysokokapacitními analytickými přístupy byla u GBM zjištěna mimo jiné dysregulace exprese některých PIWI-interagujících RNA (PIWI-interacting RNA, piRNA), krátkých jednořetězcových RNA o délce 24-32 nukleotidů, které jsou známé svojí nezastupitelnou funkcí v udržování genomové stability skrze tlumení aktivity transponovatelných elementů mechanismem vzájemné interference piRNA a cílové molekuly. piRNA byly doposud nejvíce prostudovány ve vztahu k biologii zárodečných buněk. Nedávné studie však prokázaly, že tyto molekuly hrají rovněž významnou roli v kancerogenezi mnoha nádorových onemocnění včetně GBM. Cíle diplomové práce jsou analyzovat globální expresi piRNA v GBM a nenádorových mozkových tkáních a identifikovat piRNA s významně dysregulovanou expresí, in vitro funkčně charakterizovat vybrané piRNA, které jsou asociovány s biologickým chováním GBM, a zhodnotit potenciál jejich využití jako terapeutických cílů.

Abstract:

Glioblastoma multiforme (GBM) is the most common primary brain tumor characterized by its hopeless prognosis and the survival median of 12 to 15 months from diagnosis. Biological aggressiveness of the disease and patients' length of survival often differs among individual cases. The main reason is a high heterogeneity of GBM at the cellular and molecular levels. Better understanding of intracellular signalization and relationships between molecules influencing crucial biological properties of GBM could lead to the discovery of new therapeutic approaches or new prognostic and predictive biomarkers in patients with GBM. Apart from other findings, the use of modern high-throughput analytical approaches led to the observation of dysregulated expression of PIWI-interacting RNAs (piRNAs), a group of small single-stranded RNAs with the length of 24-32 nucleotides.These molecules are known for their irreplaceable function in the maintenance of genome stability through silencing of the activity of transposable elements via the mechanism of mutual interference of piRNA and target molecule. Currently, piRNAs are described best in relation to the biology of germ-line cells. Nevertheless, recent studies proved that piRNAs play an important role in cancerogenesis of many tumors, including GBM. Aims of the study include the analysis of global expression of piRNAs in tumor and non-tumor tissues, the identification of piRNAs with a significantly dysregulated expression and the functional in vitro characterization of selected piRNAs associated with biological behaviour of GBM and evaluation of their potential use as therapeutic target.
