Induction of Cell Senescence

Abstract:

Tato bakalářská práce je věnována buněčné senescenci se zaměřením na způsoby její indukce. Nejprve je obecně popsán proces buněčné senescence, historie jejího výzkumu a její vztah s buněčným cyklem, který je regulován dráhami proteinů p16, p21, p53 a pRb. Dále jsou popsány dva typy buněčné senescence, replikativní a předčasná. V hlavní části je podrobně vysvětlen mechanismus indukce buněčné senescence a induktory, které ji spouštějí. Mezi ně řadíme epigenetické modifikátory, oxidativní stres, micro-RNA, telomery a onkogeny. U každého induktoru je popsán jeho vznik, proces, jakým působí na buňky a jeho role ve vzniku buněčné senescence. Předposlední kapitola se týká detekce buněčné senescence in vitro i in vivo, která je zde popsána pomocí tří různých metod. V závěru práce není zapomenuto ani na možné důsledky, které buněčná senescence přináší, a to v rámci běžného stárnutí, onemocnění souvisejícího s věkem a u terapeutických zákroků.

Abstract:

This Bachelor's work deals with a cellular senescence focusing on the ways of its induction. First, it generally describes the process of cellular senescence, the history of its research and its relation with the cell cycle, which is regulated by the p16, p21, p53 and pRb protein pathways. Next are described two types of cellular senescence, replicative and premature. The main part explains in detail the mechanism of induction of cellular senescence and inducers that trigger it. They include epigenetic modificators, oxidative stress, micro-RNA, telomeres and oncogenes. Each inductor is described by its origin, the process by which it acts on cells and its role in the formation of cellular senescence. The penultimate chapter is about the detection of cellular senescence in vitro and in vivo, which is described by three different methods. In the end, it is also not forgotten about the possible consequences of cellular senescence in the context of normal aging, age-related diseases and therapeutic interventions.
