Qualitative analysis of selected cleaning products by gas chromatography and their antimicrobial effect

Abstract:

Diplomová práce se zabývá problematikou dezinfekčních prostředků určené pro dezinfekci povrchů a dezinfekci pokožky. Teoretická část práce obsahuje vymezení pojmů týkající se této problematiky se zaměření na hygienickou čistotu především z hlediska mikrobiální kontaminace. Dále jsou teoreticky popsány techniky dezinfekce a použité metodiky testování dezinfekčních prostředků. Praktická část se zabývá kvalitativní analýzou komerčně dostupných přípravků a jejich porovnání s recepturou dezinfekčního přípravku dle doporučení světové zdravotnické organizace. Antimikrobiální účinnost zkoumaných dezinfekčních prostředků byla zkoumána pomocí diskové difúzní metody a ČSN EN 1040. V rámci praktické části diplomové práce byla v laboratoři zavedena jednak metodika na plynovém chromatografu a také testování podle normy zmíněné výše. Výsledky analýzy modelových vzorků dezinfekčních prostředků plynovou chromatografií ukázaly, že vhodné kombinovat více technik pomohou přesnější identifikaci většího počtu látek obsažených ve výrobcích. Všechny testované vzorky vykazovaly dezinfekční účinnost. Porovnáním výsledků plynové chromatografie a antimikrobiálního účinku vzorků bylo zjištěno, že nejvýhodnější z hlediska antimikrobiálního účinku je kombinace etanol, propan-2-olu a kvarterní amoniové sloučeniny.

Abstract:

This diploma thesis connected with the issue of disinfectants intended for surfaces and skin disinfection, respectively. The theoretical part of the work contains the definitions of terms related to this issue. It is focused on hygienic cleanliness, especially in terms of microbial contamination. Furthermore, disinfection techniques and used methods of testing disinfectants are theoretically described. The practical part deals with the qualitative analysis of commercially available products and their comparison with the disinfectant formulation prepared according to the recommendations of the World Health Organization. The antimicrobial efficacy of the investigated disinfectants was evaluated using the disk diffusion method and ČS EN 1040. The methodology of gas chromatograph and testing according to the standard mentioned above were introduced in the laboratory. The results of the analysis of model disinfectant samples by gas chromatography demonstrated that it is appropriate to combine few techniques to help accurately identify a larger number of substances contained in products. All tested samples showed disinfectant efficiency. The comparison of the gas chromatography results and the antimicrobial effect of the samples confirmed that the most advantageous composition, in terms of antimicrobial effect, is the combination of ethanol, propan-2-ol and the quaternary ammonium compound.
