Electrophoretic separations of phenolic compounds using perfluorinated carboxylic acids

Abstract:

Tato diplomová práce je zaměřena na separaci antioxidantů pomocí micelární elektrokinetické chromatografie za použití těkavých pufrů z perfluorovaných karboxylových kyselin. V prvním kroku bylo nutné zjistit elektroosmotický tok a pohyblivost micel, dále byly proměřeny jednotlivé standardy za různých podmínek. Po proměření standardů byly proměřeny nejprve jen směsi kyselin a flavonu, potom i celková směs devatenácti antioxidantů. Při žádné separaci směsi se nepodařilo rozseparovat všechny standardy, jelikož některé standardy mají stejnou pohyblivost a mohou být v jednom píku, tím pádem je v záznamu nebylo možno rozlišit. Nejlepší separace byla v 20 mM PFOA s přídavkem 10 % MeOH, v tomto případě se podařilo rozseparovat čtrnáct píků z devatenácti.

Abstract:

This diploma thesis focuses on the separation of antioxidants by micellar electrokinetic chromatography using volatile perfluorinated carboxylic acids. In the first step it was necessary to determine the electroosmotic flow and the mobility of the micelles, then the individual standards were measured under different conditions. After measuring the standards, only the mixtures of acids and flavon were first measured, then the total mixture of nineteen antioxidants. Due to similar mobilities of some standards and co-migration in one peak, it was not possible to separate all compounds in the mixture. The best separation was achiered in 20 mM PFOA with the addition of 10 % MeOH, in this case fourteen peaks from nineteen compounds were separated.
