Determination of antioxidation properties of phenolic compounds by plasmid DNA topological changes

Abstract:

Vznik volných radikálů je nevyhnutelná součást života aerobních organismů. Jejich množství v organismu je pod kontrolou antioxidačního systému. Oxidační stres, který je důsledkem nerovnováhy volných radikálů a antioxidačního systému organismu, se podílí na patogenezi řady chorob a patologických stavů. V teoretické části bakalářské práce byl charakterizován problém volných radikálů a oxidačního stresu, možnosti antioxidační ochrany organismu a vlastnosti fenolických látek. Dalším tématem teoretické části je DNA topologický test a jeho využití. V experimentální části práce bylo hlavním cílem detekovat účinky kyseliny chlorogenové na plasmidovou DNA, především inhibici oxidačniho poškozeni. Kromě protektivního účinku kyseliny chlorogenové, byl detekován také účinek prooxidační.

Abstract:

The formation of free radicals is an inevitable part of the life of aerobic organisms. Their amount in the body is controlled by the antioxidant system. Oxidative stress, which is the result of the imbalance of free radicals and antioxidant system of the body, contributes to the pathogenesis of many diseases and pathological conditions. In the theoretical part of this work the problem of free radicals and oxidative stress, antioxidant protection of the body options and features of phenolic compounds was characterized. Another issue of the theoretical part is DNA topological test and its use. In the experimental part of this work the main effects of chlorogenic acid on plasmid DNA, especially the inhibition of oxidative damage, was to detect. In addition to the protective effect of chlorogenic acid the pro-oxidant effect was also detected.
