Abstract:

Disertační práce je předkládána jako soubor komentovaných publikací, jejichž společným tématem jsou metanogenní archea, specifická společenstva podzemních zásobníků plynu. Metanogeny jsou striktně anaerobní archea s širokým spektrem výskytu napříč různými habitaty. Využívají jednoduché uhlíkaté sloučeniny za tvorby metanu jako finálního produktu jejich metabolismu. V posledních letech je také kromě environmentálně-taxono-mických studií diskutován jejich význam pro aplikace v biotechnologiích s přesahem do energetického sektoru v tzv. Power-to-Gas technologiích. Zde metan pozbývá negativní konotace potentního skleníkového plynu a nabývá nového smyslu jako produkt přebytků v procesu výroby energie z alternativních zdrojů. Disertační práce zahrnuje kvalitativní a kvantitativní studii společenstva metanogeních archea ze tří podzemních zásobníků plynu v České republice. Vzorky podzemních vod pocházející z hlubinných vrtů byly podrobeny analýze molekulárními metodami (izolace DNA, PCR, qPCR, sekvenování genu mcr-A a 16S rRNA). Výsledky poskytly náhled na složení mikrobiálních společenstev z 16 vrtů. Nejhojnějšími druhy metanogenů byly pak Methanobacterium sp., Methanothermobacter sp. a zástupci řádu Methanomicrobiales. Poznatků z první části výzkumu bylo využito jako podkladu pro unikátní pilotní experiment metanizace zásobníků in situ. Výsledkem této práce je popis složení mikrobiálního společenstva tří zásobníků s důrazem na metanogenní archea a jejich početnost. Práce zahrnuje také molekulárně biologické analýzy z provedení úspěšného pilotního experimentu metanizace v zásobníku, kde byl metan produkován při-rozeně se vyskytujícími mikroorganismy. Výsledky poukazují a potvrzují potenciál pod-zemních zásobníků plynu jako prostředí vhodného pro biotechnologickou aplikaci produkce zeleného metanu.

Abstract:

The dissertation is presented as a set of annotated publications whose common theme is methanogenic archaea, specifically the communities of underground gas reservoirs. Meth-anogens are strictly anaerobic archaea with a wide range of occurrence across different hab-itats. They utilize simple carbon compounds to produce methane as the end product of their metabolism. In recent years, in addition to environmental-taxonomic studies, their relevance for applications in biotechnology with overlap to the energy sector in so-called Power-to-Gas technologies has also been discussed. Here, methane loses its negative connotation as a potent greenhouse gas and takes on a new meaning as a surplus product in the process of energy production from alternative sources. The dissertation includes a qualitative and quantitative study of the methanogenic archaeal community from three underground gas reservoirs in the Czech Republic. The subsurface water samples from the deep wells were analysed by molecular methods (DNA isolation, PCR, qPCR, mcr-A and 16S rRNA gene sequencing). The results provided insight into the composition of the microbial communities from 16 wells. The most abundant methanogenic species were Methanobacterium sp., Methanothermobacter sp. and representatives of the order Methanomicrobiales. The find-ings from the first part of the research were used as a basis for a unique pilot experiment of in situ methanation of reservoirs. The result of this work is a description of the composition of the microbial community of the three reservoirs with emphasis on methanogenic archaea and their abundance. The thesis also includes molecular biological analyses from the conduct of a successful pilot methanation experiment in a reservoir where methane was produced by naturally occurring microorganisms of a subsurface gas reservoir. The results highlight and confirm the potential of underground gas reservoirs as a suitable pro-environment for the biotechnological application of producing green methane.
