Abstract:

Diplomová práce se zabývá hodnocením antibiotické rezistence anaerobních bakterií v čistírenském kalu. Teoretická část práce se soustředí na charakteristiku anaerobních mikroorganismů, dále na jejich specifický výskyt v čistírnách odpadních vod (ČOV) a mechanismy vzniku antibiotické rezistence. Další část se věnuje kalovému hospodářství ČOV, konkrétně technologiím, které se využívají při jeho primárním zpracování. Pozornost je také zaměřena na problematiku antibiotické rezistence v čistírenském kalu a na legislativu, která souvisí s nakládáním čistírenských kalů v čistírnách odpadních vod. Experimentální část této diplomové práce se zaměřuje na hodnocení antibiotické rezistence bakteriálních izolátů z čistírenského kalu, který byl odebrán v Centrální čistírně odpadních vod v Ostravě - Přívoze. Konkrétně byla zkoumána schopnost růstu rezistentních izolátů v aerobních a anaerobních podmínkách. Dále byly stanoveny hodnoty minimální inhibiční koncentrace ampicilinu a byla srovnána antibiotická rezistence mezi aerobními a anaerobními izoláty. Poslední metodou v experimentální časti této diplomové práce byla detekce vybraných antibioticky rezistentních genů pomocí PCR metody.

Abstract:

The thesis deals with the evaluation of antibiotic resistance of anaerobic bacteria in sewage sludge. The theoretical part of the thesis focuses on the characteristics of anaerobic microorganisms, their specific occurrence in wastewater treatment plants and the mechanisms of developing an antibiotic resistance. Next part is dedicated to the sludge management of WWTPs, specifically the technologies used in its primary processing. Attention is also focused on the issue of antibiotic resistance in sewage sludge and on legislation related to the management of sewage sludge in wastewater treatment plants. The experimental part of this thesis focuses on the evaluation of antibiotic resistence of isolates from sewage sludge collected at the Central Wastewater Treatment Plant in Ostrava - Přívoz. Specifically, the ability of the resistant isolates to grow under aerobic and anaerobic conditions. Furthermore, the values of the minimum inhibitory concentration of ampicillin were determined and antibiotic resistance was compared between aerobic and anaerobic isolates. The last method in the experimental part of this thesis was the detection of the selected antibiotic resistant genes by PCR.
