Detekce genotoxického potenciálu vzorků ovzduší eukaryotickým modelem

Abstract:

ABSTARKT Cílem práce bylo zavedení testů genotoxicity s eukaryotickými organizmy (GFP test a test s buňkami V79 hprt) a srovnání jejich citlivosti pro vzorky ovzduší s již zavedeným testem na prokaryotickém organizmu (SOS-chromotest, Escherichia coli). K odběru vzorků ovzduší byly vybrány 2 čerpací stanice pohonných hmot na území města Brna a 11 lokalit ve Zlínském kraji převážně v těsném okolí podniku DEZA a.s., který je významným emisním zdrojem PAHs. V extraktech odebraných vzorků ovzduší bylo stanoveno 16 nejvýznamnějších PAHs. Vzorky byly podrobeny třem in vitro testům genotoxicity, dvěma využívajícím eukaryotický organizmus (GFP test, test s buňkami V79 hprt), jednomu s prokaryotickými buňkami (SOS-chromotest). Principem GFP testu je detekce SOS odpovědi indikované poškozením DNA, v jehož důsledku dochází k tvorbě GFP proteinu (Green fluorescent protein). Produkce tohoto proteinu je sledována fluorimetricky. Pro detekci toxického účinku vzorku je spektrofotometricky kontrolován nárůst kultury. Test s buňkami V79 hprt je založen na detekci 6-thioguanin rezistentních mutantů, kteří přežívají v médiu s přídavkem 6-thioguaninu. Tento test je velice časově, materiálově a prostorově náročný. SOS-chromotest využívá detekce SOS odpovědi pomocí spektrometrického stanovení produkce -galaktosidázy, která indikuje poškození DNA. Test byl použit jako srovnávací test pro nově zavedené testy na eukaryotických systémech. Důležitým výsledkem mé práce je zavedení dvou nových testů genotoxicity na eukaryotických buňkách do laboratoří výzkumného centra RECETOX, včetně jejich optimalizace. Oba tyto testy prokázaly vysokou citlivost k účinkům testovaných vzorků. GFP test je navíc rychlým screeningovým testem lze jej tedy doporučit pro zařazení do baterie testů genotoxicity. Většina vzorků volného ovzduší vykazovala významnou genotoxicitu. Výsledky SOS-chromotestu dobře korelují s obsahem PAHs stanoveným chemickým rozborem testovaných vzorků. Abstract:

ABSTRACT The aim of this study was the introduction of genotoxicity tests on eucaryotic organisms (GFP test and test on V79 hprt cells) and a comparison of their sensitivity with the test on a procaryotic organism (SOS-chromotest, Escherichia coli) for air samples. As sampling sites two petrol stations in the urban area of Brno were chosen and eleven locations in the Zlín district, mainly in the close neighborhood of DEZA a.s. Company, which is a significant source of PAHs. 16 most important PAHs were detected in the extracts of taken samples. Three in vitro genotoxicity tests were performed, two of them based on eukaryotic organisms (GFP test, test on V79 hprt cells), the other one on prokaryotic organism (SOS-chromotest). The principle of the GFP test is detection of SOS response indicated by DNA damage, which induces formation of GFP protein (Green fluorescent protein). The production of GFP protein is detected by the use of fluorimeter, detection of toxic effect of the sample the growth of the culture detected. The test on V79 hprt cells is based on the detection of 6-thioguanin resistant mutants, which survive in the medium with 6-thioguanin additions. This test is very demanding on time, material and place. The SOS-chromotest takes advantage of SOS response detection by the help of spektrofotometric determination of -galaktosidaze enzyme, which indicates DNA damage. An important result of my work is introduction and optimalization of two new genotoxicity assays on eukaryotic cells to the laboratories of RECETOX centre. Both these assays manifest high sensitivity to the effects of tested samples. In addition, GFP test is a rapid screening test and it can be recommended to be included to the battery of genotoxicity assays. The majority of the air samples showed a significant genotoxicity. The results of the SOS-chromotest are well correlating with the content of PAHs determined by chemical analysis of tested samples.
