Transcript analysis of genes related to grapevine resistance

Abstract:

Tato práce je zaměřena na optimalizaci metody měření exprese genů biosyntetických drah rostlinného hormonu kyseliny salicylové, který zprostředkovává signalizaci v procesu rostlinné imunity známém jako sysetmic aquired resistance, pomocí kvantitativní real time PCR v rostlinách Révy vinné. Cílem práce je vybrat a optimalizovat metodu izolace RNA z listů rostlin Révy vinné, provést kvantitativní real time PCR a zároveň vybrat vhodné referenční geny, které by mohly být použity v této metodě. V práci bylo zjištěno, že vhodnou metodou pro izolaci RNA je metoda využívající CTAB extrakční pufr a že vhodnými referenčními geny pro metodu PCR jsou geny UBC a PEP.

Abstract:

This thesis is focused on the optimalization of the method of measuring expression of genes involved in biosynthetic pathways of phytohormone salicylic acid, which is crucial member of signalisation pathways in systemic acquired resistance process in plants, in grapevine plants. The method of quantitative real time PCR will be used in the thesis. The goal of the thesis is to choose and optimize suitable method of isolation of RNA from grapevine leaves, perform the quantitative real time PCR and choose suitable reference genes for this method. The thesis discovers that CTAB extraction buffer-based method is the suitable for RNA isolation and that genes UBC and PEP are suitable reference genes for PCR method.
