Hormonal profiles and social structure: using non-invasive methods for estimating the level of steroid hormones in the study of house mouse populations under semi-natural conditions and in behavioural experiments

Abstract:

V rámci projektu zaměřeného na sledování dynamiky tvorby a vývoje sociální struktury u domácích myší v polopřirozených podmínkách byly porovnány hladiny metabolitů dvou klíčových hormonů, kortikosteronu a testosteronu, v trusu myší chovaných odděleně ve standardních laboratorních podmínkách i skupinově v polopřirozených chovech. Mým cílem bylo srovnat hladiny těchto hormonů mezi dvěma poddruhy, Mus musculus musculus a M. m. domesticus, i mezi pohlavími, posoudit případný vliv kontaktu se standardním oponentem i vliv vstupu do sociálního prostředí, zaznamenat dynamiku hladin obou hormonů během celého období experimentu v polopřirozených chovech a konečně zhodnotit jejich změnu po ukončení experimentu. Výsledky ukázaly, že samice mají obecně vyšší hladiny kortikosteronu než samci a že tento rozdíl je zvlášť patrný u M. m. domesticus. Co se týče testosteronu, potvrdila se jeho vyšší hladina u samců musculus, a to jak v izolaci, tak v sociálním prostředí arén. Dále byla potvrzena hypotéza, že kontakt s neagresivním samcem inbredního kmene A/J vyvolává u samců musculus zvýšení produkce kortikosteronu i testosteronu, kdežto u M. m. domesticus nemá tento kontakt na hladinu testosteronu žádný vliv, v případě kortikosteronu dokonce překvapivě dochází k jeho výraznému poklesu. Vstup do prostředí polopřirozených chovů v počáteční fázi vytváření sociální struktury vyvolal zvýšení tvorby obou hormonů, jejichž hladiny pak zůstaly víceméně stabilní po celou dobu experimentu, s výjimkou samců domesticus, u nichž byl zaznamenán stálý pokles jak kortikosteronu, tak testosteronu. To naznačuje odlišnou dynamiku tvorby sociální hierarchie u obou poddruhů. Ukončení polopřirozených chovů a opětovná izolace u samců vedla k velmi rychlému poklesu hladin obou hormonů na bazální úroveň zjištěnou před začátkem experimentu. Tyto závěry ukazují na významné rozdíly mezi oběma blízce příbuznými taxony a tím i na riziko zobecňování výsledků získaných u jednoho poddruhu na celý druh.

Abstract:

Within a project focused on the study of the dynamics of establishing and evolution of the social structure in house mice under semi-natural conditions, I have compared levels of two key hormones, corticosterone and testosterone, in faeces of mice kept in isolation under standard lab conditions as well as in semi-natural enclosures. I aimed at comparing levels of both hormones between two mouse subspecies, Mus musculus musculus and M. m. domesticus, and between males and females, assessing potential influence of a contact with a standard opponent and an impact of entering the social environment, monitoring of the dynamics of the hormones during the whole experiment in semi-natural conditions, and, finally, assessing the changes in the hormonal levels after termination of the experiment. The results showed that females tended to have higher levels of corticosterone than males, this difference being especially apparent in M. m. domesticus. In testosterone, higher levels in musculus than in domesticus were confirmed both in isolation and in social conditions of the enclosures. Furthermore, I confirmed the hypothesis that the contact with a non-aggressive male of the A/J inbred strain causes an increase of both corticosterone and testosterone production in musculus males whereas no effect on testosterone was revealed in M. m. domesticus – in the latter, strikingly, the corticosterone level was even substantially reduced. After launching the semi-natural enclosures, the process of establishing the social structure had elicited an increase in the production of both hormones and their levels have then stayed more or less stable during the whole experiment, except of domesticus males displaying a continuous decrease of corticosterone and testosterone as well, suggesting the different dynamics of formation of the social structure in the two subspecies. Closing the enclosures and subsequent re-isolation has led in males to a rapid decrease of the hormones to basal levels established before launching of the experiment. These conclusions highlight significant differences between the two closely related taxa and hence risk of generalizing the results obtained in a single subspecies to the whole species.
