Determination of Selected Fatty Acids in Human Blood Obtained by Dried Spot Technique

Abstract:

Tato práce se zabývá možností stanovení vybraných mastných kyselin v lidské krvi (získané technikou suché kapky) metodou plynové chromatografie s hmotnostní detekcí. Práce je zaměřena na přípravu vzorku před analýzou a sleduje vliv některých faktorů, které ovlivňují kvantitativní výsledky. Byly testovány různé techniky zavádění vnitřního standardu ke vzorku, stabilita vybraných mastných kyselin ve vzorku suché kapky krve při pokojové teplotě, 4 °C a -20 °C s přídavkem a bez přídavku antioxidačního činidla, butylovaného hydroxytoluenu. Z analytických parametrů metody byly určeny linearita a nepřesnost v sérii.

Abstract:

This thesis deals with the possibility of determining selected fatty acids in human blood (obtained by the dried spot technique) by the method of gas chromatography with mass detection. This thesis is focused on sample preparation before analysis and the influence of some factors affecting quantitative results. Different techniques of introducing an internal standard to the sample were tested, the stability of selected fatty acids in a sample of dried blood spot at room temperature, 4 °C, and -20 °C with and without the addition of the antioxidant agent, butylated hydroxytoluene. Linearity and imprecision were determined from the analytical parameters of the method.
