Wastewater treatment plants as a potential source of ATB resistant microorganisms

Abstract:

Diplomová práce je zaměřena na čističky odpadních vod jako potenciální zdroj ATB rezistentních mikroorganismů. Rešeršní část práce se zabývá charakteristikou čistíren odpadních vod a jejich mechanickým i biologickým čištěním. Kapitola zahrnuje i charakteristiku nitratačních a nitritačních bakterií, které jsou využívané při biologickém čištění. V následující kapitole je věnována pozornost antimikrobiálním látkám, jejich rozdělení a mechanismu účinku. Další část práce se zabývá rezistencí bakterií na antimikrobiální látky, mechanismus rezistence k beta-laktamovým antibiotikům a význam mobilních genetických elementů, které mají hlavní roli v šíření rezistence. V experimentální části dochází k zaměření na čističku odpadních vod Lučina v blízkosti Žermanické přehrady, kde byly odebrány a testovány vzorky z aktivační nádrže, dosazovací nádrže a rybníku. Ze vzorků byla izolovaná DNA. Následně proběhla detekce ARGs pomocí PCR. Na závěr u detekovaných genů byla provedena real-time PCR. Byl sledován, jak rozdíl množství vyskytujících se genů v nádrži, tak i rozdíl časového odběru vzorků. Poslední část je zaměřena na korelaci mezi koncentrací antibiotik stanovenou HPLC v odtoku a výskytem ARGs.

Abstract:

The master thesis is focused on wastewater treatment plants as a potential source of ATB resistant microorganisms. The research part of the work deals with the characteristics of wastewater treatment plants and their mechanical and biological treatment. The chapter also includes the characteristics of nitratating and nitritating bacteria, which are used in biological treatment. In the following chapter, attention is paid to microbials, their distribution and mechanism of action. The next part of the work deals with the resistance of bacteria to antimicrobials, the mechanism of resistance to beta-lactam antibiotics and the importance of mobile genetic elements, which play a major role in the spread of resistance. In the experimental part focus on the wastewater treatment plant Lučina near the Žermanická dam, where samples were taken and tested from nitrification tank, sedimentation tank and ponds. DNA was isolated from the samples. Subsequently, ARGs were detected by PCR. Finally, real-time PCR was performer on the detected genes. Different numbers of genes present in the tanks were monitored, as well as different time sampling. The last part is focused on the correlation between the concentration of antibiotics determined by HPLC in the effluent and the occurrence of ARGs.
