Determination of daidzin, genistin and glycitin in feed using HPLC

Abstract:

Fytoestrogeny jsou širokou skupinou přirozeně se vyskytujících sekundárních rostlinných metabolitů. Za jejich hlavní zdroj jsou považovány sója a sójový bob z rostliny Glycine max, která je řazena do čeledi bobovitých (Fabeceae). Isoflavonové fytoestrogeny se vyznačují svou strukturní podobností k 17-β-estradiolu, díky čemuž jsou schopny se svými fenolytickými kruhy navázat na estrogenní receptory a být tak estrogenními agonisty či antagonisty. Z tohoto důvodu jsou dnes předmětem mnoha studií, které se jimi zabývají jak z hlediska možné hormonální terapie, tak z hlediska jejich působení jako estrogenní disruptory indukující různé choroby. Bakalářská práce se věnovala zavedením metody pro stanovení β-glykosidů i aglykonů v rámci jedné analýzy za pomoci vysokoúčinné kapalinné chromatografie (HPLC). Díky vyvinuté metodě určené pro HPLC analýzu lze detekovat isoflavony, které byly obsaženy ve vzorku. V rámci bakalářské práce byla také vyvíjena nová metoda pro zpracování vzorku krmiva. Nově tedy postup přípravy a zpracování vzorků zahrnuje jejich vystavení vůči působení ultrazvuku. V rámci metody jsme schopni identifikovat píky isoflavonů na chromatogramu a na základě ploch píků určit jejich koncentraci ve vzorku. Nejefektivnější postupem přípravy vzorků se ukázala metoda B, kde za použití 75% MeOH jako rozpouštědla byly stanoveny největší koncentrace glykosylovaných isoflavonů i příslušných aglykonů.

Abstract:

The phytoestrogens are a large group of naturally appearing secondary plant metabollites. A soy bean (Glycine max) that belongs to the family of legumes (Fabeceae) is considered to be their main source. Their structural similarity with 17-β-estradiol is characteristic for isoflavone phytoestrogens. Thanks to that they are able to bind to estrogen receptors with their phenolitic rings and so be estrogenic agonists and antagonists. Because of this fact they are nowadays a topic of many studies. In these studies phytoestrogens are considered as a possible alternative for hormonal therapy and as estrogen disruptors inducing various diseases. This thesis deals with implementation of a method that can determine β-glycosides and aglycones by using high performance liquid chromatography (HPLC). The method based on HPLC analysis can detect isoflavones that are contained in a sample. In the bachelor thesis a new method for a sample prepartion has been developed as well. The new step of a sample preparation includes their exposure to the ultrasound. The method can determine isoflavones contained in a sample, indentify their chromatographic peaks and also based on a peak area determine their concentration. The most effective method of a sample preparation turned out to be the method B that includes using 75% MeOH as a solvent. With this method of preparation the highest concentrations of glycosylated isoflavones and also their aglycones were determined.
