Analysis of selected markers in head and neck squamous cell carcinoma

Abstract:

Spinocelulární karcinomy hlavy a krku představují heterogenní skupinu nádorů spojených s agresivním chováním, vysokou četností rekurence a vysokou mortalitou. Pro zlepšení prognózy pacientů s HNSCC je zapotřebí hledat nové diagnostické, prognostické a prediktivní markery, určit jejich funkci v karcinogenezi a pochopit dynamiku změn jejich hladiny v průběhu terapie. Tato práce se zabývá efektem cisplatiny, docetaxelu a zinečnatých iontů na fenotypové projevy a chování nádorových buněčných linií FaDu a Detroit 562. Metodou MTT byly stanoveny hodnoty padesátiprocentní inhibiční koncentrace pro všechny testované látky a obě nádorové linie a tyto koncentrace byly využity v dalších testech. Byl určen vliv testovaných látek na expresní profil genů vybraných napříč procesy karcinogeneze, na koncentrace aminokyselin a metalothioneinu, na migrační schopnost a invazivitu buněk a jejich schopnost zakládat nové kolonie. Bylo zjištěno, že testované látky významně ovlivnily expresi genů zapojených do angiogeneze a apoptózy a že použití cytostatik a zinečnatých iontů mělo odlišný efekt na koncentraci metalothioneinu, některých aminokyselin a také na schopnost buněk zakládat kolonie. Odlišné efekty zinku v porovnání s cytostatiky naznačují, že by mohl podporovat chemorezistenci a progresi HNSCC.

Abstract:

Head and neck squamous cell carcinomas represent a heterogeneous group of tumours characterized by aggressive behaviour, high rate of recurrence and mortality. To improve the prognosis of patients with HNSCC, it is necessary to look for new diagnostic, prognostic and predictive markers, to clarify their function in carcinogenesis and to understand the dynamics of changes in their levels during treatment. The goal of this thesis is to determine the effect of cisplatin, docetaxel and zinc ions on phenotypic manifestations and behaviour of FaDu and Detroit 562 cancer cell lines. Half maximal inhibitory concentrations of all treatments were determined for both cancer cell lines and these concentrations were used in other tests. The effect of treatment on gene expression, amino acids and metallothionein concentrations, cell migration, invasiveness and the ability of these cell lines to establish new colonies was assessed. We found out that tested substances significantly affected the expression of genes involved in angiogenesis and apoptosis and that used cytostatics and zinc ions had different effect on the concentration of metallothionein, some amino acids and also on the ability of cells to establish new colonies. These results suggest that zinc compared to cytostatics could support tumour chemoresistance and progression of HNSCC.
